Ο καταρροϊκός πυρετός ή νόσος της κυανής γλώσσας (Bluetongue disease, BT) είναι ιογενής λοιμώδης νόσος των προβάτων και άλλων οικόσιτων και άγριων μηρυκαστικών που μεταδίδεται με έντομα του γένους Culicoides. Ανήκει στην κατηγορία των νοσημάτων που πρέπει να δηλώνονται άμεσα στο Διεθνές Γραφείο Επιζωοτιών, διότι διασπείρεται ταχέως με τη μετακίνηση των Culicoides και προκαλεί μεγάλες οικονομικές απώλειες στην κτηνοτροφία των μηρυκαστικών των χωρών όπου εμφανίζεται. Οφείλεται στον ιό του καταρροϊκού πυρετού (Bluetongue Virus, BTV) που ανήκει στο γένος Orbivirus (οικογένεια Reoviridae). Είναι ιός αντιγονικά πολύμορφος και συγκροτεί ορολογική ομάδα 24 οροτύπων, οι οποίοι έχουν διαφορετική γεωγραφική κατανομή. Η πρόληψη του νοσήματος στηρίζεται στο σύστημα επιτήρησης εισόδου και κυκλοφορίας στελεχών του ιού σε μία χώρα, στον έλεγχο του πληθυσμού των Culicoides, αλλά και στους εμβολιασμούς. Οι κλασικές μέθοδοι ανίχνευσης και τυποποίησης στελεχών του ιού (ενοφθαλμισμός σε εμβρυοφόρα αυγά ή σε κυτταροκαλλιέργειες για την απομόνωση του ιού, ανοσοφθορισμός ή ανοσοϋπεροξειδάση και DAS-ELISA για ανίχνευση της ορολογικής ομάδας και εξουδετέρωση με γνωστούς αντιορούς έναντι των 24 οροτύπων για την τυποποίηση του οροτύπου) είναι διαδικασίες χρονοβόρες, δαπανηρές, απαιτούν εξειδικευμένο προσωπικό, ενώ αδυνατούν να διακρίνουν τα άγρια από τα εμβολιακά στελέχη. Για να ξεπεραστούν τα προβλήματα αυτά, ορισμένοι ερευνητές ανέπτυξαν διάφορα πρωτόκολλα RT-PCR προτείνοντας τα για την αντικατάσταση των κλασικών μεθόδων. Όμως, μέχρι την έναρξη της παρούσας έρευνας, τα πρωτόκολλα RT-PCR που είχαν προταθεί για την ανίχνευση της ορολογικής ομάδας παρουσίαζαν διακυμάνσεις στην ευαισθησία και το εύρος ανίχνευσης. Παράλληλα τα πρωτόκολλα RT-PCR που είχαν αναπτυχθεί για την τυποποίηση του οροτύπου αφορούσαν οροτύπους που δεν είχαν εμφανισθεί σε χώρες της Μεσογείου και τη Μέση Ανατολή και επομένως δεν ήταν δυνατό να χρησιμοποιηθούν για την τυποποίηση των οροτύπων (BTV-1, -4, -8, -9 και -16) που κυκλοφορούν σε χώρες της Ευρώπης. Σκοπός της παρούσας έρευνας ήταν να επιλυθεί το πρόβλημα της ταχείας τυποποίησης των στελεχών του ιού BTV, αλλά και να μελετηθούν μοριακά τα στελέχη που απομονώθηκαν στην Ελλάδα, ώστε να συγκεντρωθούν στοιχεία για την επιζωοτιολογική διερεύνηση του καταρροϊκού πυρετού. Για το λόγο αυτό, σε ένα πρώτο βήμα απομονώθηκαν με κλασικές μεθόδους στελέχη του ιού από διάφορες επιζωοτίες και αναπτύχθηκε μία μοριακή δοκιμή ( ημι-ένθετη RT-PCR) με βάση το γονίδιο του κοινού αντιγόνου (VP7) της ορολογικής ομάδας, ενώ στη συνέχεια με βάση το γονίδιο του ειδικού αντιγόνου (VP2) του κάθε οροτύπου, αναπτύχθηκαν πρωτόκολλα RT-PCR με ειδικούς εκκινητές που τυποποιούσαν τα στελέχη των οροτύπων BTV-1, -4, -9 και -16. Για τη μοριακή μελέτη του γενωμικού τμήματος L3 έγινε εξακρίβωση της αλληλουχίας των νουκλεοτιδίων ελληνικών στελεχών του ιού BTV και ακολούθησε η φυλογενετική τους ανάλυση, ώστε να προσδιοριστεί η προέλευση των στελεχών που προκάλεσαν επιζωοτίες καταρροϊκού πυρετού.

Four different RT-PCR methods were also developed to specifically detect the genome segment 2 of BTV-1, -4, -9, and -16 serotypes. To evaluate the polyvalency of the RT-PCR methods, a total of 59 field isolates were used. Along with these, 14 additional strains from other countries and the 24 BTV reference strains representing all known serotypes were also included in our investigation. The specificity of the tests was evaluated using 9 representative reference strains of the closely related Orbiviruses (EHDV, AHSV and EEV) and negative samples (tissue culture cells and sheep blood). The semi-nested RT-PCR proved to be polyvalent and specific, capable of detecting all isolates except this of serotype 7. The method was also sensitive, since a virus titre of 0.178 TCID50/0.1 ml could be detected. Moreover, preliminary results using blood samples from serial bleedings of experimentally inoculated sheep and cattle with BTV-8, indicated that this method could be used for detecting the virus from the 3rd to 27th day post inoculation. It could be approved that this test is a fast, useful and cost-effective tool for BTV monitoring and certifying ruminants as BTV-negative, before movement. Concerning the four different RT-PCR methods for serotyping, 17 different primer pairs were evaluated. Some primer pairs were not able to detect any BTV strain, while two other primer pairs, designed for detecting BTV-4 and -9, also detected strains belonging to serotypes of the same nucleotype. Additionally, one primer pair proved to be inefficient to detect all BTV-16 field isolates. Finally, 5 primer pairs were selected as the most efficient ones for specifically detecting each of the BTV-1, -4, -9 and -16 serotypes. Therefore, they can be used as a great tool for rapid diagnosis in taking control measures (eradication programs / vaccinations) and in epidemiological studies. Molecular characterization and phylogenetic analysis based on genome segment 3 of 14 Greek BTV isolates, representing all identified serotypes, was also performed. Due to lack of enough genetic information for this genome segment of BTV-1, isolates from Nigeria, Malaysia and India were also analyzed. Full-length nucleotide sequences of genome segment 3 were obtained using a new sequencing strategy based on the ligation of an “anchor-primer”. All sequenced segments 3, along with all available full length published sequences were used to obtain phylogenetic inferences with “Maximum Likelihood” analysis. Results showed that the Greek isolates belong to two different lineages, representing “North American-South.African” and “Australasian” topotypes respectively. The Greek BTV-4 isolates were almost identical irrespectively of the year of isolation, host species and geographic location. They belonged to the “North American-South.African” lineage and were closely related to BTV isolates originating from Corsica in 2000. Thus the Greek BTV-4 strains belong to the group of BTV viruses circulating in the Mediteranian Basin. In the contrary, the BTV-1, -9 and -16 isolates belonged to the “Australasian” lineage and formed two different monophyletic clades. Greek BTV-1 isolates were almost identical and related to BTV isolates originating from India. BTV-9 and -16 clustered together showing very small evolutionary distances indicating a possible reassortment. These isolates formed a cluster that was separate from the other isolates of the “Australasian” lineage indicating a different geographic origin that could be in the Middle East. From the alignment of the resulting nucleotide and amino acid sequences it was proved that segment 3 and the coding protein VP3, is very conserved among all isolates suggesting that significant functional and structural constrains exist.
