A topographic and comparative study of scrapie histopathological lesions and the presence of PrPsc in the central nervous system of sheep and goats
Abstract
The aim of this work was to conduct a topographic and comparative study of scrapie histopathological lesions and PrPsc deposition in the Central Nervous System (CNS) of sheep and goats in relation to age, presence or absence of clinical signs and polymorphisms of the PrPc genotype. 65 sheep (30 with clinical signs of scrapie and 35 “clinically healthy”) and 73 goats (8 with clinical signs of the disease and 65 “clinically healthy”), aged 1-10 years were used in this study. The animals originated from Greek flocks in which scrapie had been diagnosed with histopathological and biochemical methods. EDTA-treated blood was collected from all animals for genotyping. The brains were routinely fixed and one cervical spinal cord segment as well as eight coronal slices of the brain (medulla oblongata at the level of the obex and the trapezoid body, middle of the pons, mesencephalon, cerebellum and mammilary body, hypophyseal infudibulum and rostral to corpus callosum levels) were selected for hi ...
