Πρωτεωμική ανάλυση πλάσματος υπό συνθήκες έντονου στρες

Περίληψη

Το Σπάρταθλον (η κάλυψη της απόστασης 246 km σε λιγότερο από 36 h) χρησιμοποιήθηκε ως μοντέλο έντονου σωματικού στρες για τη μελέτη πρωτεϊνικών μεταβολών στο πλάσμα των αθλητών στην αφετηρία (Αθήνα - Φάση Ι), στον τερματισμό (Σπάρτη – Φάση ΙΙ) του αγώνα, καθώς και 48h μετά την ολοκλήρωση του αγώνα (Φάση – ΙΙΙ). Το πλάσμα των αθλητών αναλύθηκε με δισδιάστατη ηλεκτροφόρηση (2-DE) και οι διαφορικά εκφρασμένες πρωτεΐνες ταυτοποιήθηκαν με ιοντισμό με εκρόφηση από μήτρα μέσω λέιζερ (MALDI-TOF) φασματομετρία μάζας (MS). Τα αποτελέσματά μας ταυτοποιήθηκαν με ανοσοαποτύπωμα κατά Western και με βιοχημικές αναλύσεις. Συνολικά πενήντα-μια πρωτεΐνες βρέθηκαν διαφορικά εκφρασμένες μεταξύ της εκκίνησης, του τερματισμού και δύο μερών μετά την ολοκλήρωση του αγώνα. Από αυτές οι τριάντα σχετίζονταν με τη φλεγμονή , ενώ οι υπόλοιπες ήταν αντιοξειδωτικές, αντιπηκτικές, ή σχετίζονταν με τον μεταβολισμό και με τη μεταφορά το σιδήρου. The Spartathlon race (brisk walking a distance of 246 km in less than 36 h) was employed as a model of severe physical stress to investigate proteomic alterations in the plasma of athletes at the start (Athens – Phase I) and finish (Sparta– Phase II) of the race, as well as 48h after the race (Post– Phase III). The athletes’ plasma was analysed by 2D gel electrophoresis (2-DE) and the differentially expressed proteins were identified by matrix-assisted laser desorption ionisation-time of flight (MALDI-TOF) mass spectrometry (MS). Our results were confirmed by Western blot and biochemical analyses. Overall fifty-one proteins were differentially expressed between the starting point, the finishing line and two days after the end of the race. Thirty proteins were involved in the inflammatory process, while the rest concerned anti-oxidant, anti-coagulant and metabolic processes as well as iron transport.
