ων. Το θεωρητικό μέρος της διατριβής αποτελειται από 4 κεφάλαια. Στο κεφάλαιο 1 περιγράφονται οι μοναδικές ιδιότητες των νανοσωματιδίων και ιδιαίτερα των νανοσφαιριδίων χρυσού. Τα νανοσφαιρίδια χρυσού παρουσιάζουν έντονο κόκκινο χρώμα που οφείλεται στο συντονισμό των επιφανειακών πλασμονίων, στα 520 nm. Με σύζευξη των νανοσφαιριδίων με βιομόρια είναι δυνατή η αξιοποίησή τους στην ανάλυση νουκλεϊκών οξέων. Στο κεφάλαιο περιγράφονται επίσης ορισμένες εφαρμογές τους. Το κεφάλαιο 2 αναφέρεται στις ταινίες ξηρών αντιδραστηρίων με τη χρήση έγχρωμων νανο/μικροσφαιριδίων και οι εφαρμογές τους στην ανάλυση των νουκλεϊκών οξέων. Το κεφάλαιο 3 αναφέρεται στη φαρμακογενετική και τη φαρμακογενωμική, αντικείμενο των οποίων είναι η συσχέτιση των πολυμορφισμών που παρουσιάζονται σε γονίδια που συμμετέχουν στο μεταβολισμό φαρμάκων, με την απόκριση του οργανισμού στα φάρμακα. Στο κεφάλαιο 4 περιγράφονται τα γονίδια-στόχοι, ο ρόλος και η σημασία τους. Το πειραματικό μέρος του διδακτορικού αποτελείται από 5 κεφάλαια. Στο κεφάλαιο 5 περιγράφεται η οργανολογία, τα αντιδραστήρια και οι μέθοδοι που χρησιμοποιήθηκαν στο πειραματικό μέρος. Στο κεφάλαιο 6 περιγράφεται η ανάπτυξη ταχείας μεθόδου προσδιορισμού πολυμορφισμών σε γονίδια που επιδρούν στο μεταβολισμό φαρμάκων. Η μέθοδος βασίζεται στην ανίχνευση των προϊόντων της γονοτυπικής αντίδρασης PEXT με ταινία ξηρών αντιδραστηρίων. Η προτεινόμενη μέθοδος περιλαμβάνει εκθετική ενίσχυση με PCR, της περιοχής του γονιδίου που περιέχει το σημειακό πολυμορφισμό, δύο αντιδράσεις PEXT χρησιμοποιώντας αλληλο-ειδικούς εκκινητές, και δοκιμασία με ταινία ξηρών αντιδραστηρίων μιας ζώνης δοκιμασίας. Η ταινία ξηρών αντιδραστηρίων χρησιμοποιήθηκε για την ανίχνευση βασικών πολυμορφισμών στα γονίδια CYP2D6, CYP2C19 και TPMT και συνολικά πραγματοποιήθηκαν 76 γονοτυπικοί προσδιορισμοί. Τα αποτελέσματα ήταν σε πλήρη συμφωνία με αυτά που προέκυψαν έπειτα από αλληλούχιση DNA. Στο κεφάλαιο 7 περιγράφεται η αξιοποίηση έγχρωμων μικροσφαιριδίων πολυστυρενίου ως ιχνηθετών σε ταινίες ξηρών αντιδραστηρίων. Τα σφαιρίδια πολυστυρενίου διατίθενται στο εμπόριο τροποποιημένα με ποικιλία χαρακτηριστικών ομάδων και η σύζευξη τους με ειδικά ολιγονουκλεοτίδια πραγματοποιείται ομοιοπολικά με τη βοήθεια ειδικών αντιδραστηρίων και κατάλληλα τροποποιημένων ολιγονουκλεοτιδίων. ...............................

This doctoral thesis is focused on the exploitation of dry-reagent rapid tests (dipsticks) in nucleic acid analysis that combines the highly specific recognition properties of oligonucleotides with the unique optical properties of gold nanoparticles. The dry-reagent rapid tests are suitable for “point-of-care” applications, for the detection of amplified nucleic acid products, as well as for the detection of polymorphisms/mutations of genes. The theoretical part of this thesis consists of 4 chapters. Chapter 1 describes the unique properties of nanoparticles and especially those of gold nanoparticles (ΑuNPs). The bright red color of ΑuNPs is due to the plasmon resonance peak at 520 nm. The functionalization of ΑuNPs with biomolecular recognition motif provides flexibility for a variety of applications.
