The role of polymorhism of HER-2/neu ongoprotein and of cytokinew in breast cancer development

Abstract

PURPOSE: Alterations of c-erbB-2 (neu or HER-2) proto-oncogene have been associated with carcinogenesis and poor prognosis of breast cancer. A single nucleotide polymorphism (SNP) at codon 655 resulting in a G to A transition (Ile655Val) in the transmembrane domain-coding region of this gene has been associated with an increased risk of breast cancer, particularly among younger women. The frequency of the Val allele was highly variable between ethnic groups. This study aims to determine the prevalence of HER-2 genotype and its association with breast cancer in the population of Thrace consisting of Greek Christians and Greek Muslims. The prognostic significance of serum levels of TGF-β1, TNF-a, IL-6 and extracellular domain of HER-2 (sHER-2) in breast cancer was evaluated and their correlation with other examinations and clinicopathological aspects and HER-2 polymorphism was also investigated.
