FORMULATION AND IN-VITRO, IN-VIVO RELEASE STUDIES OF DRUGS FROM SEMISOLID DOSAGE FORMS
Abstract
SEVERAL SEMISOLID DOSAGE FORMS CONTAINING THEOPHYLLINE, OR THEOPHILLINE SODIUM GLYCINATE (TS6) OR SULINDAL WERE PREPARED TO DETERMINE THE IN-VITRO RELEASE OF THE DRUGS. THE GENERAL RANK ORDER FOR THE IN-VITRO RELEASE OF THE DRUGS WAS FOUND TO BE: GELS>MICROEMULSIONS>EMULSION GELS>EMULSION O/W>EMULSION W/O. THE SELECTED GEL CONTAINING THEOPHYLLINE 1% HAD NO EFFECT ON THE TREATMENT OF PSORIASIS. TSG WAS RELEASED FASTER IN-SITU FROM A GEL CONTAINING 2% TSG THAN FROM EMULSIONS O/W AND W/O. PENETRATION ENHANCERS SUCH AS DMSO, DMF AND UREA AT 5% AND 10%CONCENTRATION LEVELS HAD NO SIGNIFICANT EFFECT IN ENHANCING THE IN-SITU RELEASE OF TSG. THE IN-SITU RELEASE RATE OF THE DRUG WAS INCREASED AS THE CONCENTRATION LEVEL OF TSG WAS INCREASED FROM 2% TO 5% AND 10% WITHIN THE GEL BASE. THE PERCUTANEOUS ABSORPTION OF THEOPHYLLINE FROM THE SELECTED GEL (WITH 10% TSG) WAS STUDIED IN RABBITS AND IT WAS FOUND CMAX=3.28MG/ML, TMAX=10HRS, AUCO-24=49. 7MG.HR/ML.
