Η παρούσα εργασία παρουσιάζει τα αποτελέσματα της ενοποίησης μεθόδων από διαφορετικά γνωστικά αντικείμενα, με στόχο την κατανόηση των επιδράσεων της σωματιδιακής ύλης στην ανθρώπινη υγεία. Για το σκοπό αυτό δείγματα σωματιδιακής ύλης από οχήματα διαφορετικής τεχνολογίας συλλέχθηκαν ως υδάτινα εναιωρήματα, με την βοήθεια ενός συστήματος εμπλουτισμού αερολυμάτων (VACES). Η τεχνική αυτή οδηγεί στη συλλογή σωματιδιακής ύλης, διατηρώντας αναλλοίωτα τα φυσικοχημικά χαρακτηριστικά των σωματιδίων. Δείγματα σωματιδίων συλλέχθηκαν από ένα βενζινοκίνητο όχημα προδιαγραφής Euro 3 και από δύο οχήματα diesel που τηρούσαν τις προδιαγραφές Euro 2 και Euro 4. Το όχημα προδιαγραφής Euro 2 εφοδιάστηκε με συμβατικό καύσιμο diesel και στη συνέχεια με 100% βιοντίζελ από σογιέλαιο. Το όχημα προδιαγραφής Euro 4 μετρήθηκε και εναλλακτικά με φίλτρο αιθάλης. Η παρούσα εργασία παρουσιάζει τα αποτελέσματα της ενοποίησης μεθόδων από διαφορετικά γνωστικά αντικείμενα, με στόχο την κατανόηση των επιδράσεων της σωματιδιακής ύλης στην ανθρώπινη υγεία. Για το σκοπό αυτό δείγματα σωματιδιακής ύλης από οχήματα διαφορετικής τεχνολογίας συλλέχθηκαν ως υδάτινα εναιωρήματα, με την βοήθεια ενός συστήματος εμπλουτισμού αερολυμάτων (VACES). Η τεχνική αυτή οδηγεί στη συλλογή σωματιδιακής ύλης, διατηρώντας αναλλοίωτα τα φυσικοχημικά χαρακτηριστικά των σωματιδίων. Δείγματα σωματιδίων συλλέχθηκαν από ένα βενζινοκίνητο όχημα προδιαγραφής Euro 3 και από δύο οχήματα diesel που τηρούσαν τις προδιαγραφές Euro 2 και Euro 4. Το όχημα προδιαγραφής Euro 2 εφοδιάστηκε με συμβατικό καύσιμο diesel και στη συνέχεια με 100% βιοντίζελ από σογιέλαιο. Το όχημα προδιαγραφής Euro 4 μετρήθηκε και εναλλακτικά με φίλτρο αιθάλης. Συνολικά, μετρήθηκαν πέντε διαμορφώσεις οχημάτων που οδήγησαν στη συλλογή πέντε αντίστοιχων δειγμάτων σωματιδίων. Σωματιδιακή ύλη από τις πέντε διαμορφώσεις οχημάτων συλλέχθηκε επιπρόσθετα με τον παραδοσιακό τρόπο σε φίλτρα. Στα δείγματα αυτά προσδιορίστηκε η ποσότητα στοιχειακού άνθρακα (EC), οργανικού άνθρακα (OC), υδατοδιαλυτού οργανικού άνθρακα (WSOC), μετάλλων και ανόργανων ιόντων και επιπλέον προσδιορίστηκανεπιμέρους οργανικά συστατικά (χοπάνια, στεράνια, οργανικά οξέα, πολυαρωματικοί υδρογονάνθρακες). Ο προσδιορισμός του οξειδωτικού στρες που προκαλεί το κάθε δείγμα ποσοτικοποιήθηκε με τη βοήθεια δύο αναλύσεων, μιας χημικής μεθόδου (κατανάλωση ουσίας Dithiothreitol) και μιας in vitro μεθόδου με την οποία προσδιορίζεται η παραγωγή δραστικών ριζών οξυγόνου (ROS) από τα κύτταρα. Τα δείγματα σε εναιωρήματα εγχύθηκαν ενδοτράχεια στους πνεύμονες ποντικών σε δύο επίπεδα δόσης (50 μl και 100 μl). Μετά από 24 ώρες οι ποντικοί υποβλήθηκαν σε βρογχοεκπλύσεις (BAL) με σκοπό τον προσδιορισμό οξείων φλεγμονών και αιματολογικών μεταβολών. Τα αποτελέσματα έδειξαν ότι τα δείγματα σωματιδιακής ύλης προκαλούν μέση αλλά στατιστικά σημαντική αντίδραση του οργανισμού τωνποντικών, η ένταση της οποίας εξαρτάται από το όχημα – πηγή της σωματιδιακής ύλης. Οι αντιδράσεις του οργανισμού παρουσιάζουν ισχυρή συσχέτιση με διάφορα οργανικά και ανόργανα συστατικά όπως το βένζο(a)ανθρακένιο, το χρυσένιο, τα Mn, Fe και Cu,ορισμένους βαρείς PAH αλλά και με το δυναμικό της σωματιδιακής ύλης να ερεθίζει τα κύτταρα με αποτέλεσμα αυτά να παράγουν ROS. Τελικά, σχεδιάζονται ποιοτικά καμπύλες «αντίδραση-δόση» για τις πέντε διαμορφώσεις οχημάτων που μετρήθηκαν. Τα αποτελέσματα αποκαλύπτουν ότι η φλεγμονώδης αντίδραση του οργανισμού δεν είναι γραμμική συνάρτηση της ποσότητας της δόσης με αποτέλεσμα η παρατηρούμενη τοξικότητα να εξαρτάται από το επίπεδο της δόσης. Η μέθοδος που αναπτύχθηκε επιτρέπει την εξαγωγή κάποιων αρχικών συμπερασμάτων σε σχέση με την επίπτωση των σωματιδίων από διαφορετικές πηγές στη λειτουργία κυττάρων και οργανισμών. Προφανώς, επιβεβαίωση και διευκρίνιση των συμπερασμάτων αυτών απαιτεί περαιτέρω μελέτη, με επέκταση ή/και επανάληψη των μετρήσεων σε συνδυασμούς οχημάτων/καυσίμων. Η παρούσα εργασία παρέχει ωστόσο τα εργαλεία για μια τέτοια διερεύνηση. Στο πλαίσιο αυτό, τα κύρια συμπεράσματα που εξήχθησαν μπορούν να συνοψιστούν στα ακόλουθα σημεία: 1. Χρησιμοποιήθηκαν πέντε διαφορετικές μεθοδολογίες εκτίμησης της τοξικότητας και η σύνοψη των αποτελεσμάτων δείχνει τα εξής: • Η μέθοδος DTT είναι ευαίσθητη στο οργανικό μέρος της σωματιδιακής ύλης. Έτσι, όταν τα αποτελέσματα εκφράζονται ανά μονάδα μάζας της σωματιδιακής ύλης τα οχήματα ιεραρχούνται με βάση την τοξικότητα τους ως εξής: B100 Euro 2> D Euro 4> D Euro 4+> D Euro 1> G Euro 3. Από την άλλη μεριά, αν τα αποτελέσματα εκφραστούν ανά οχηματοχιλιόμετρο, η σειρά αλλάζει και γίνεται: D Euro 1> B100 Euro 2> D Euro 4> G Euro 3> D Euro 4+. Τα οχήματα B100 Euro 2 και D Euro 4 βρίσκονται ψηλά στην ιεραρχία ανεξάρτητα από τον τρόπο έκφρασης της τοξικότητας. • Η μέθοδος ROS είναι εξαιρετικά ευαίσθητη στα μεταβατικά μέταλλα που βρίσκονται στη σωματιδιακή ύλη. Η μέθοδος αυτή αναδεικνύει το όχημα G Euro 3 ως το πιο τοξικό ανεξάρτητα από τον τρόπο έκφρασης των αποτελεσμάτων (είτε ανά μάζα, είτε ανά οχηματοχιλιόμετρο). Ακολουθούν τα οχήματα D Euro 1 και B100 Euro 2 με μεγάλη διαφορά στην τοξικότητα. Η τοξικότητα της σωματιδιακής ύλης που εκπέμφθηκε από τις δύο διαμορφώσεις του οχήματος Honda Accord δεν ήταν ανιχνεύσιμη με τη συγκεκριμένη μέθοδο. • Η μέθοδος BAL χρησιμοποιήθηκε για την ποσοτικοποίηση της οξείας φλεγμονώδους αντίδρασης του οργανισμού και της βλάβης των ιστών. Αναφορικά με την οξεία φλεγμονώδη αντίδραση, δύο ήταν τα οχήματα που οδήγησαν σε στατιστικά σημαντικά αποτελέσματα: το G Euro 3 και το D Euro 1. Από την άλλη μεριά, βλάβη σε ιστούς προκάλεσαν όλα τα οχήματα diesel, όταν στους ποντικούς δόθηκε η υψηλή δόση των 100 μl. Μάλιστα, η μεγαλύτερηβλάβη προκλήθηκε από το όχημα D Euro 4. Με τη δόση των 50 μl τα οχήματα που έδωσαν στατιστικά σημαντικά αποτελέσματα ήταν τα G Euro 3 και D Euro 1. • Η μέθοδος FMT έδωσε ενδείξεις για σημαντική τοξικότητα του οχήματος G Euro 3. • Η μέθοδος του κρυοστάτη έδωσε ενδείξεις για τοξικότητα του οχήματος με βιοντίζελ. 2. Από τα παραπάνω αποτελέσματα, ο χαρακτηρισμός του πιο τοξικού συνδυασμού οχήματος/καυσίμου μπορεί να αποδοθεί στη διαμόρφωση G Euro 3. Ακολουθούν τα οχήματα B100 Euro 2, D Euro 1 και D Euro 4. Ο λιγότερο τοξικός συνδυασμός οχήματος/καυσίμου είναι το όχημα D Euro 4+. 3. Η εργασία αυτή παρείχε τα εργαλεία για τη διεξαγωγή πειραμάτων εκτίμησης τοξικότητας με αποτελέσματα αρκετά ενθαρρυντικά. Εντοπίστηκαν ενώσεις που φαίνεται να συμμετέχουν κυρίαρχα στους μηχανισμούς τοξικότητας (Fe, Mn, Cu, βένζο(a)ανθρακένιο, χρυσένιο και βαρείς πολυαρωματικοί υδρογονάνθρακες). Ηδιαφορετική φύση των ουσιών αυτών (μέταλλα, υδρογονάνθρακες) αντικατοπτρίζουν τουλάχιστον δύο βασικούς μηχανισμούς τοξικότητας. Τέλος, αποκαλύφθηκε η αδυναμία εκτίμησης της τοξικότητας ενός δείγματος από μία μοναδική δόση. 4. Η επανάληψη των πειραμάτων με μεγαλύτερο αριθμό μετρήσεων και ποντικών και η επέκταση των μετρήσεων σε διαφορετικούς συνδυασμούς οχήματος/καυσίμου, αφενός θα ενισχύσει τη γνώση σχετικά με τους μηχανισμούς τοξικότητας, αφετέρου θα διευκολύνει την ανεύρεση των πραγματικών μηχανισμών τοξικότητας που ενεργοποιούνται από τη σωματιδιακή ύλη στα κύτταρα και στους οργανισμούς.

With respect to environmental effects, PM is characterised as an absorbing aerosol (Menon et al. 2002). Absorbing aerosols heat the air, alter regional atmospheric stability and vertical motions, and affect the largescale circulation and hydrologic cycle with significant regional climate effects. Although epidemiology is able to identify the versatile impact of particulate matter on human health, it is not able, as an observational science, to draw conclusions about the biological mechanisms that lead to the various diseases observed. This role isundertaken by toxicology. The knowledge and training of a mechanical engineer guarantee the representative collection of particle samples, which will constitute the toxic substance under examination. This study intends to develop a methodology for the assessment of the toxicity of particles emitted from vehicles, in order to shed light to the underlying mechanisms, through which particles threaten human health.2. Methodology Three passenger cars were selected for testing, including a Euro 3 gasoline (G Euro 3), and one Euro 2 and one Euro 4 (D Euro 4) diesel cars. The Euro 2 diesel was powered by 100% (B100) soybean-derived biodiesel and is designated as “B100 Euro 2”. Also, some alternative configurations were tested to increase the range of technologies examined. In an alternate configuration for the Euro 2 diesel vehicle, the oxidation catalyst originally employed was removed and the vehicle was powered with conventional (fossil) diesel to represent Euro 1 technology (D Euro 1). Also, further to its original configuration, the Euro 4 car was retrofitted with a diesel particle filter (DPF) to study the toxicity of emissions at the low emission levels imposed by the DPF (D Euro 4+). In this way, these five configurations promised particulate emissions with significant variability in their physicochemical characteristics. Theoretically, this diversity in particle characteristics will increase the contrast between the toxicity mechanisms, thus making them more obvious. The measurement protocol used to measure the five configurations, included five tests: four driving cycles and a steady-state test at 90 km h-1. The cycles that were employed involved the legislated New European Driving Cycle and the three Artemis cycles (André 2004). PM mass samples were collected on filters (T1, Q1), in accordance with the legislated process (1999/96/EC), for chemical speciation and gravimetric analysis. ACondensation Particle Counter (TSI Inc.) counted the total number of particles, along with a Dekati Mass Monitor. For the measurement of the non-volatile particles, an Electrical Low Pressure Impactor was employed, which was sampling downstream of a thermodenuder set at 250 °C to remove volatile and semi-volatile particle components. A scanning mobility particle sizer (SMPS, Model 3936L, TSI Inc.) replaced the CPC in steady-speed tests to measure the mobility size distribution. The SMPS operated on a sheath over sample flow rate of 10/1 l min–1 and a scan time of 90 s. Along with the sampling setup described above, an Versatile Aerosol Concentration Enrichment System (VACES) was collecting aerosol in water suspensions by means of an impinger (Biosampler, SKC Inc.). The system flowrates were set at 100.5 lpm and 6 lpm for the major and the minor flowrate, respectively. The enriched aerosol was also collected on filters (T2, Q2) that were sampling in parallel with the biosampler. The employed setup allows the exact determination of the water suspension PM contentthrough the chemical analysis of the filters. The filters that were sampling upstream of the VACES would be used for the the detailed determination of the chemical composition of PM emitted from each vehicle, while extractions of PM from the filters would be usedfor toxicological analysis. The waterborne suspensions from the VACES would also be used for toxicological analysis. The identification of the underlying toxicity mechanisms would be attempted by correlating the results of toxicological tests and those of the chemical analysis. This correlation is enabled through the agreement obtained by comparing the results of chemical analysis between upstream (T1, Q1) and downstream of VACES filters (T2, Q2). For this correlation, the enrichment factor of VACES is needed. The enrichment factor that was used was equal to 16 (resulting from the employed flowrates of VACES). The chemical analysis applied on the filters are shown below: Teflon Filters (T1) • Water-soluble organic carbon (WSOC) • Ions speciation: [chloride (Cl-), nitrate (NO3-), phosphate (PO43-), sulphate (SO42-), sodium (Na+), ammonium (NH4+), and potassium (K+)] Quartz Filters (Q1) • Elemental carbon (EC) • Organic Carbon (OC) • Water soluble trace elements: (Na, Mg, Al, K, Ca, Ti, V, Cr, Be, S, Mn, Fe, Co, Cu, Zn, Mo, V, Sr, Ba, Cd, Pt, Pb and Ni) • Organic compounds: polycyclic aromatic hydrocarbons (PAH), hopanes, steranes, n-alkanes, and organic acids) • Extraction with water for determination of cellurar production of reactive oxygen species (ROS) Teflon Filters (T2) • Water-soluble organic carbon (WSOC) • Ions speciation: [chloride (Cl-), nitrate (NO3-), phosphate (PO43-), sulphate (SO42-), sodium (Na+), ammonium (NH4+), and potassium (K+)] Quartz Filters (Q2)• Elemental carbon (EC) • Organic Carbon (OC). The toxicological assays performed with the water suspensions are the following:• Chemical method of Dithiothreitol (DTT) consumption rate • Bronchoalveolar Lavage (BAL) on mice (Balb/cj) for measurement ofpolumorphonuclear white cells (PMNs) as a marker of inflammation • Bronchoalveolar Lavage (BAL) on mice for measurement of tissue damage by determining protein concentration in BAL fluids • Non-invasive imaging technique (Fluorescence molecular tomography) appliedon mice to identify inflammation of the lung using the fluorescent substance ProSense750 • Invasive imaging method applied on mice using a cryostat and the same fluorescent substance. The results of the analysis above as well as the correlations between them are referredbelow. 3. Results The measured vehicles meet the respective specifications regarding PM emissions. It should be noted that yhe use of biodiesel results in substantial reduction of PM. The vehicles with diesel fuel, but without diesel particle filter (DPF), emit particles in theorder of 1013 km-1 on average throughout the protocol. The gasoline vehicle, as well as the vehicle with the DPF emit substantially lower levels of airborne particles compared to the previous ones, with the corresponding value being equal to 1011 km-1. The obtained size distributions show that the particle volume has decreased approximately 20%, when shifting from Euro 1 to Euro 4. On the other hand, the number of non-volatile particles emitted by these two vehicles is almost the same. The size distribution corresponding to the vehicles with biodiesel is quite interesting, as a nucleation mode occurs with a peak at 20 nm. The proper handling of the distributions based on the aerodynamic and the mobility diameters leads to the calculation of the fractal dimension (DF) of the particles. The three vehicles with diesel fuel, without a DPF, are characterized by particles with an average DF equal to 2.5 and a reference effective density equal to 0.85 g cm-3. The other two vehicles emit more spherical and compact particles (DF=3). The calculated density profiles can lead to more accurate calculation of the number of non-volatile particles, as they were measured by the ELPI. The comparison between the number of non-volatile particles and the total number of airborne particles reveals that the particles emitted from the G Euro 3 and the D Euro 4+ vehicles are mainly consisting of volatile compounds. The chemical analysis revealed two basic results. Firstly, the determination of particulate matter by means of chemical analysis agrees well (R2=1, slope=1) with the legislated gravimetric analysis. The chemical determination of PM was performed by summing the elemental carbon, the organic mass (determined by the organic carbon after using a correction factor ranging from 1.3-1.5, depending on the vehicle and the corresponding ratio of WSOC/OC) and the ions. Secondly, the PM determined bychemical analysis of filters upstream and downstream of VACES also correlates quite well (R2=0.8, slope=1). This result enables the correlation between the results of toxicological analysis (waterborne suspensions downstream of VACES) and the results of chemical analysis in filters upstream of VACES, in order to highlight the underlying mechanisms and the triggering chemical groups. Particulate matter emitted from vehicles D Euro 4 and D Euro 1 consists of elemental carbon at a perecentage higher than 55%. This rate is below 15% regarding the rest vehicles. In the case of the gasoline vehicle, 50% of PM is ions (70% of these ions is sulfates). Chemical analysis showed that the D Euro 1 vehicle emits the highest levels of PAH, while the only diesel vehicle emitting heavy PAHs is the biodiesel vehicle.However, the gasoline vehicle emits higher levels of heavy PAHs compared to the biodiesel vehicle. The DPF filter is proved to be very effective in reducing the majority of the chemical constituents measured. The toxicological analysis led to variable results, apparently identifying more than one mechanisms of toxicity. The summary of the toxicity results are presented below: 1. The DTT substance is intensely consumed by vehicles D Euro 1 and B100 Euro2. 2. The cellular ROS production is intensely triggered by the particulate matteremitted from the gasoline vehicle. 3. The number of neutrophils (PMNs) is increased in the cases of D Euro 1 and G Euro 3 vehicles, in the case of the high dose (100 μl). 4. On the other hand, D Euro 4 is the vehicle causing the most extensive tissue damage. 5. The imaging method using cryostat gives indications of possible increased toxicity of the biodiesel vehicle compared to the gasoline vehicle. The rest of the samples were not measured. 6. The organisms’ reactions are strongly correlated with the active redox elements, such as Fe, Mn, and Cu. 7. The P, S and Pb elements are strongly correlated with the organisms’ reaction. As far as Pb is concerned, the oxidative stress has been explained by the inhibition of functional sulfonyl groups (Mudipalli 2007) from various antioxidant enzymes. On the other hand, P and S are not known to be active redox elements. In conclusion, their statistical correlation with the biological reactions exists, either due to an unknown alternative mechanism or due to their internal relationship with the elements Mn and Fe. 8. Finally, toxicity is highly correlated with some PAHs [benzo(a)anthracene and chrysene] and the categories of medium and heavy PAHs. 4. Conclusions: The developed methodology can be easily implemented, while no special knowledge is required. The results contribute to the identification of the underlying toxicity mechanisms as well as to the overall toxicity assessment of PM. The metals Fe, Mn and Cu, but also organic compounds (especially PAHs) showed a strong correlation with the results of toxicological analysis. Biodiesel leads to increased PAHs emissions and also to more compact particles. Soot filters promise to reduce the majority of harmful chemical substances, while it is demonstrated that the biological response of an organism can not be inferred by using a single dose. The construction of the entire "dose-response" curveis a safer and more realistic approach.
