Η εργασία αποκαλύπτει τη δυναμική της προστατευτικής δράσης του σιδήρου, σε πειραματικά μοντέλα δερματικής λεϊσμανίασης, ιδιαίτερα όταν οι πειραματικές συνθήκες προσεγγίζουν τις πραγματικές συνθήκες μετάδοσης του παρασίτου. Ο «ανθεκτικός» φαινότυπος που αναπτύσσουν οι μύες BALB/c στους οποίους χορηγείται σίδηρος έναντι των παρασίτων L. major συνδέεται με τα σταθερά αυξημένα επίπεδα της οξειδωτικής έκρηξης που μέσω των παραγομένων ενεργών οξειδωτικών ριζών επιτρέπουν τη συνεχή καταστροφή των παρασίτων. Επιπλέον, οι ενεργές οξειδωτικές ρίζες φαίνεται να λειτουργούν και ως δεύτερα μηνύματα τροποποιώντας το αρχικό δυναμικόμικροπεριβάλλον μέσα στο οποίο ενεργοποιούνται τα φαγοκύτταρα και τα αντιγονοπαρουσιαστικά κύτταρα του ξενιστή. Η «ανθεκτικότητα» που εμφανίζουν οι μύες στους οποίους χορηγείται σίδηρος στην επαναμόλυνση αποτελεί μια επιπλέον ισχυρή ένδειξη ότι ο σίδηρος τροποποιεί τους ανοσιακούς μηχανισμούς προς την κατεύθυνση της ανοσιακής «προστασίας» Thl.Τα πειραματικά μοντέλα που μελετήθηκαν μπορούν να αποτελέσουν χρήσιμα εργαλεία για την περαιτέρω μελέτη των σχέσεων μεταξύ των αποθηκών σιδήρου του ξενιστή, των οξειδωτικών ριζών και των μακροπρόθεσμων σηματοδοτικών λειτουργιών τους. Επιπλέον οι μελέτες αυτές έχουν αξία γιατί μπορούν να πραγματοποιηθούν και σε ιστούς όπου ο παρασιτισμός δεν έχει εμφανή κλινικά συμπτώματα -όπως είναι η καταστροφή των ιστών- και επομένως προσομοιάζει με τον ασυμπτωμαπκό παρασιτισμό κατά τον οποίο το δέρμα των ασυμπτωματικών φορέων αποτελεί «αποθήκη» του παρασίτου. Τέλος, τα πειραματικά μοντέλα που μελετήθηκαν στην παρούσα εργασία μπορούν να δώσουν σημαντικές πληροφορίες για τη σχέση της παραγωγής ενεργών οξειδωτικών ριζών και της έκβασης του παρασιτισμού με μακροπρόθεσμες εφαρμογές διαγνωστικής και θεραπευτικής αξίας στον άνθρωπο.

Làshmania spp. are parasites establishing long term interactions in mammals -such as rodents, canids, and humans. These eukaryotic parasites are transmitted by the phlebotomine sand fly in the skin of mammals and can provoke various and complex parasitic processes ranging from asymptomatic parasitism to more or less severe clinical manifestations. Using the inoculation of a high dose of stationary phase L. major promastigotes in the footpad of BALB/c mice, we previously established that iron delivery, at time points surrounding the parasite inoculation, was sufficient to result in a footpad lesion of reduced severity. In this model, it was possible to establish that the full protective status relies on dynamic processes that are disrupted by the delivery of an antioxidant molecule, the DiPhenylenelodonium chloride (DPI). Thus the efficient parasite containment inside phagocytic leukocytes and the absence of tissuedamaging processes could be attributed, at least in part, (a) to the observed increase in the oxidative burst and (b) the ROS-dependent regulation of sustained though dynamic tissue- protective processes.In this study, iron-loaded BALB/c mice were re-inoculated, at a distant site (intra-dermal re-inoculation of 1000 metacyclic promastigotes in the contra-lateral ear) and were found resistant contrary to non treated mice. We considered that NF-xB activation could be a key process involved in the iron-induced protective and resistance to re-inoculation status. Therefore we asked whether the sustained “protective” phenotype observed in iron- loaded BALB/c mice was associated with NF-xB activation at late time points post parasite inoculation. When analyzed by EMSA at week 8 post the first parasite inoculation (just before the second parasite inoculation) and at weeks 8, 12 post the second parasite inoculation, a consistent NF-xB DNA-binding activity was detected only in the nuclear extracts prepared from the leukocytes present in the DLN of iron-loaded mice. Addition of polyclonal antibody selected for binding activity to the NF-xB p65 subunit resulted in a shift in NF-xB complexes, indicating that the NF-xB-DNA complex consisted of the p65 subunit. In order to investigate whether the increased oxidative burst, the NF-xB activation and the TCR dependent- proliferation potential of CD3+ lymphocytes could be coupled processes, cells from the DLN and spleen of non-treated and iron-loaded mice were recovered at weeks 8 and 12 post the second parasite inoculation. It was observed that TCR -mediated T-lymphocyte proliferation was enhanced and higher number of IFN γ- positive CD4+ T-cells were found in cell populations recovered from iron-loaded mice.Results from this study provide insights on complex signaling pathways underlying the protective and resistant status induced by iron in BALB/c mice. The ROS-dependent activation of the NF-κΒ, assessed ex vivo, could be, at least partly, a process allowing the onset and long term maintenance of tissue-protective processes in the parasite-inoculated site. These processes are responsible for the renewed presence in the DLN of balanced populations that not only prevent the parasite progeny to grow but prevent the skin to be damaged.
