Ένας βασικός στόχος του Προγράµµατος Ανάλυσης του Ανθρώπινου Γονιδιώµατος είναι η ταυτοποίηση και λειτουργική ανάλυση νέων γονιδίων. Στη χρωµοσωµική περιοχή 10q23.3-25.1 χαρτογραφούνται πολλά µη ταυτοποιηµένα και αρκετά σχετιζόµενα µε γενετικές ασθένειες, γονίδια. Ένα από αυτά είναι το NEURL, το ανθρώπινο οµόλογο του γονιδίου neuralized (neur) της Drosophila, που χαρτογραφείται στη θέση 10q25.1, την περιοχή µε τις πιο συχνές χρωµοσωµικές ανωµαλίες σε καρκινικά αστροκυττώµατα. Το neur της Drosophila όπως και τα γονίδια Notch (N), Delta (Dl), Enhancer of split [E(spl)], big brain (bib), mastermind (mam) και almondex (amx), ανήκει στα νευρογόνα γονίδια. Εκφράζεται σε πληθώρα ιστών σε πολλά αναπτυξιακά στάδια και συµµετέχει σε διαδικασίες διακυτταρικής επικοινωνίας, όπου κύριος ρυθµιστής είναι ο διαµεµβρανικός υποδοχέας Notch. Με βάση τα χαρακτηριστικά του δροσοφιλικού neur, διατυπώθηκε η υπόθεση ότι το οµόλογο γονίδιο του ανθρώπου, NEURL, πιθανόν να εµπλέκεται στις διαδικασίες καθορισµού της κυτταρικής τύχης στο νευρικό σύστηµα και ότι η απενεργοποίηση του σχετίζεται µε την δηµιουργία καρκινικών νευροεκτοδερµικών όγκων. Προκειµένου να διευκολυνθεί η λειτουργική διερεύνηση του neuralized στα θηλαστικά, αποµονώσαµε και µελετήσαµε το οµόλογο γονίδιο στον ποντικό, Neurl. Λόγω της περιορισµένης πληροφορίας που υπήρχε σχετικά µε τη λειτουργία της δροσοφιλικής πρωτεΐνης Neur, αλλά και για να εκµεταλλευτούµε το πρόσφορο λειτουργικό σύστηµα της Drosophila, θεωρήσαµε σκόπιµο να µελετήσουµε παράλληλα και τα δύο γονίδια neur και Neurl, ώστε η διαλεύκανση της λειτουργίας του neur να αξιοποιηθεί για τη διερεύνηση της λειτουργίας του γονιδίου των θηλαστικών. Οι πρωτεΐνες Neuralized του ποντικού και της Drosophila, έχουν συντηρημένη αρχιτεκτονική δομή καθ’ όλο το μήκος των µορίων τους. Τα κύρια κοινά χαρακτηριστικά των πρωτεϊνών αποτελούν δύο επαναλαμβανόμενες περιοχές, NEUZ1 και NEUZ2, άγνωστης λειτουργίας και ο δάκτυλος ψευδαργύρου RING, που αποτελεί χαρακτηριστική δομική περιοχή λιγάσης ουβικουϊτίνης Ε3. Πειράµατα προσδιορισµού του προτύπου έκφρασης έδειξαν ότι το γονίδιο Neurl είναι ενεργό από τα αρχικά στάδια της εμβρυογένεσης του ποντικού, στο νευρικό και στηρικτικό σύστημα, στα αναπτυσσόμενα άκρα και στα επιθήλια των εσωτερικών και αισθητηρίων οργάνων. Παροδική έκφραση χιμαιρικών πολυπεπτιδίων Neur ή Neurl, αποτελούμενων από την πράσινη φθορίζουσα πρωτεΐνη (GFP) συνδυασμένη µε ολόκληρα µόρια ή υποπεριοχές τους, σε κυτταρικές σειρές θηλαστικών καθώς και πειράματα κατευθυνόμενης υπερέκφρασης στη Drosophila, έδειξαν ότι οι πρωτεΐνες ανιχνεύονται στην κυτταρική µμεμβράνη και πιθανότατα σε ενδοκυττωτικά κυστίδια. Από το συνδυασμό βιοχημικών και ανοσο-ιστοχηµικών μεθόδων, επίσης, διαπιστώθηκε ότι η σύνδεση των µορίων Νeuralized στη μεμβράνη του κυττάρου και στα κυστίδια δεν είναι άμεση αλλά οφείλεται σε αλληλεπιδράσεις µε διαµεµβρανικές πρωτεΐνες ή άλλους παράγοντες της κυτταροπλασµατικής περιφέρειας. Για την σωστή υποκυτταρική τοποθέτηση των πρωτεϊνών Neurl ή Neur, είναι ικανό και αναγκαίο ένα τμήμα της πρωτεΐνης που εκτείνεται από το αµινοτελικό άκρο μέχρι την περιοχή NEUZ1. Η λειτουργική συγγένεια των πρωτεϊνών διαπιστώθηκε µε επιπρόσθετα πειράματα στο σύστημα-μοντέλο της Drosophila. Κατευθυνόμενη υπερέκφραση του διαγονιδίου neur ή Neurl είχε παρόμοια φαινοτυπικά αποτελέσματα σε ενήλικα άτομα. Παρατηρήθηκε διακοπή του ορίου του φτερού και πάχυνση των φλεβών, φαινότυποι που χαρακτηρίζουν την απώλεια λειτουργίας του σηµατοδοτικού µηχανισµού Notch. Επίσης παρατηρήθηκε καταστολή της έκφρασης του γονιδίου E(spl)m8, ενός µεταγραφικού στόχου του Notch. Όπως ήταν αναμενόμενο λόγω της εξελικτικής απόστασης, η επίδραση του θηλαστικού πολυπεπτιδίου στο μονοπάτι Notch ήταν µικρότερης έντασης. Αναλυτικότερη λειτουργική μελέτη της πρωτεΐνης της Drosophila έδειξε ότι παίζει σημαντικό ρόλο τόσο στην ενίσχυση της ικανότητας του δεσµευτή Delta (Dl) να ενεργοποιεί τον υποδοχέα Notch (Ν) όσο και στην μετέπειτα ενδοκύττωση, in cis, και αποικοδόµησή του πρώτου. Επιπλέον, δείχθηκε ότι το Dl συμπαρασύρει σε ενδοκύττωση και την εξωκυττάρια περιοχή του Notch (NEC). Ακόμη, παρατηρήθηκε ότι ο δάκτυλος RING είναι υπεύθυνος για την ενδοκυτταρική µμεταφορά του συμπλέγματος Dl-NEC. Αντίθετα, αυτή η περιοχή δεν συµµετέχει στην ενίσχυση της επαγωγικής ικανότητας του δεσµευτή Delta. Τα παραπάνω αποτελέσματα υποδεικνύουν ότι η πρωτεΐνη Νeuralized παίζει σηµαντικό ρόλο στις διαδικασίες διαφοροποίησης λειτουργικά ισοδύναμων κυττάρων και στην υιοθέτηση της διαφορετικής αναπτυξιακής τους πορείας.

In order to contribute to the elucidation of the functional role of neuralized in mammals, we isolated and analyzed the structure and the expression of themouse ortholog, Neurl. Furthermore, we decided to analyze in parallel, both the insect and the mouse molecules using the well-studied Drosophila experimental model system. This approach would offer us the opportunity to obtain particularly useful information regarding the biological role of the insect polypeptide and at the same time, extrapolate this information to the mammalian homolog. Mammalian and insect neuralized proteins share extensive structural similarities through-out the entire molecule. The most recognizable similarities are two neutralized specific sequence repeats, NEUZ1 and 2, of unknown function and a carboxy-terminal RING Zinc finger, characterizing proteins with E3 ubiquitin ligase activity. Expression profile determination in embryo and adult mouse tissues, suggested that Neurl is active from the earliest embryonal developmental stages in neural and skeletal system, in the developing limbs and in epithelial cell layers of sense and internal organs. Transient expression in mammalian cell lines of GFP-tagged neuralized polypeptides (using either full length or truncated molecules) as well as targeted over-expression of either Neur or Neurl in Drosophila tissues, showed that both proteins are localized in the cytoplasmic side of the plasma membrane and possibly, in endocytotic vesicles. Biochemical experiments and immunofluorescence microscopy observations suggested that the association of neuralized protein with the membrane is possibly due to interactions with transmembrane proteins or with other molecules of the cytoplasmic periphery. Furthermore, it was shown that the amino-terminal part of the neuralized protein extended up to the first NEUZ domain, is necessary and sufficient for its subcellular localization. The functional similarities of the insect and the mammalian Νeuralized were further corroborated with extensive functional experimentation in the Drosophila model system. Targeted over-expression of a neur or Neurl transgenes in Drosophila embryos resulted in similar phenotypic consequences, i.e.: loss of Notch function in adults as manifested by cut wings and vein thickening. Down-regulation of E(spl)m8, a transcription target gene of the Notch signaling pathway, at the wing margin was also observed. Possibly due to the evolutionary distance between the two homologs, the phenotypic effects of the mammalian transgene were weaker than those observed with the Drosophila neur. Detailed functional analysis of the Drosophila gene was undertaken in order to elucidate the exact function of Neuralized protein in Notch signaling and to get as much information as possible concerning the role of the murine polypeptide. Thus, it was shown that Neur plays a significant role in enhancing the ability of Delta ligand to activate Notch receptor, as well as in subsequent endocytosis, in cis, and post translational degradation of the former. Moreover, Delta endocytosis is followed by Notch extracellular region co-internalization. Furthermore, the Neur RING domain is required for Dl internalization but is dispensable for Dl signaling activity. The potent modulatory effect of Neur on Dl activity makes Neur a good candidate for establishing signaling asymmetries and thus, different developmental fates within cellular equivalence groups.
