Τα πρωτεϊνικά ικριώματα τα οποία σχηματίζονται από αυτοοργανωμένα πεπτίδια μπορούν να χρησιμοποιηθούν ως εναλλακτικά των πρωτεϊνών της εξωκυττάριας μήτρας (ECM) και θα μπορούσαν να βρουν εφαρμογή στην αναγεννητική ιατρική. Στο πρώτο κεφάλαιο της παρούσας διατριβής ένα πεπτίδιο που περιέχει την αμυλοειδή αλληλουχία NSGAITIG η οποία έχει ανακτηθεί από το μίσχο της πρωτεΐνης, ίνας του αδενοϊού ανασχεδιάστηκε για την κατασκευή τρισδιάστατων (3D) ικριωμάτων ανθρώπινου ιστού και μοντέλων κυτταρικής καλλιέργειας in vitro. Εν συνεχεία αυτή η ακολουθία μελετήθηκε έτσι ώστε να διαμορφωθεί ένα εντεκαπεπτίδιο το οποίο περιέχει το μοτίβο RGD στο αμινοτελικό άκρο και ένα κομβικό αμινοξύ όπως η κυστεΐνη στο καρβοξυτελικό άκρο, έτσι ώστε να σχηματιστεί η αλληλουχία RGDSGAITIGC ως τελικό προϊόν. Εναλλακτικά, στο δεύτερο κεφάλαιο, φωτοευαίσθητα κατάλοιπα όπως η τυροσίνη ή η τρυπτοφάνη σχεδιάστηκαν για να εισαχθούν στο καρβοξυτελικό άκρο ώστε να δώσουν τις αλληλουχίες RGDSGAITIGY και RGDSGAITIGW. Αυτά τα κατάλοιπα μπορούν να πυροδοτήσουν ένα φαινόμενο πολυφωτονικού πολυμερισμού (MPP/MPA) μέσω της χρήσης της τεχνικής της Direct Laser Writing (DLW) με απώτερο στόχο την εφαρμογή σε υψηλά ανταποκρινόμενες βιολογικές θεραπείες, όπως η αναγέννηση του μυοκαρδίου. Εν συνεχεία, στο τρίτο κεφάλαιο, τα πεπτιδικά παράγωγα τα οποία χαρακτηρίστηκαν προηγουμένως, μελετήθηκαν ως υποστρώματα στη θεραπεία βλαστοκυττάρων. Επιπλέον τροποποιημένα εμβρυϊκά βλαστοκύτταρα μελετήθηκαν για κυτταρική προσκόλληση και πολλαπλασιασμό. Επιπροσθέτως, η μελέτη των ρεολογικών ιδιοτήτων όλων των υποστρωμάτων και η περαιτέρω αξιολόγηση της λειτουργικής έγχυσης μελετήθηκε με σκοπό την προοπτική θεραπευτικής χρησιμοποίησής τους in situ.

Fibrous protein-like scaffolds deriving from self-assembling (S.A.) peptides are a valid alternative to extracellular matrix (ECM) proteins and are deemed to be an effective contrivance towards cell-based regenerative medicine with attainable perspectives. In the first chapter of this thesis work, a peptide containing an amyloidogenic sequence (NSGAITIG) previously retrieved in the adenovirus fiber shaft protein was projected for bio-fabrication of 3D tissue scaffolds and in vitro cell culture models. Thereafter, this sequence was studied to be incorporated as a S.A. leading core in an endecapeptide containing the cell-attachment motif RGD at the N-terminus and carrying a key residue such as cysteine to give RGDSGAITIGC (RGD_C). In pertinence to hard tissue engineering, the understanding of the cell interactions with the biomaterial and consequently the evaluation of cell-attachment and proliferation are the basic steps for the long-term revolutionary objective of developing cell-based photo-structured 3D “scaffold-on-scaffold” constructs derived from combining top-down stereolithography and bottom-up bio-fabrication typical of self-assembling peptides. Alternatively, photosensitive residues such as tyrosine or tryptophan were designed to be inserted at the C-terminus to give RGDSGAITIGY and RGDSGAITIGW. These residues could potentially trigger a Multi Photon Polymerization/Ablation (MPP/MPA) phenomenon through the use of the Direct Laser Writing (DLW) technique in a method that encounters the needs of using a high-responding biological therapy such as in myocardial regeneration and consequently, a biomaterial fully degradable. In chapter 2, with regard to fabricate precise 3D scaffolds, the aforementioned photosensitive S.A. peptide networks were irradiated both by a non-linear femtosecond laser and a KrF 1-photon pulsed laser. This latter, was used also to examine the production of cross-linked moieties through Laser-Induced Fluorescence technique. De facto, the change of the peak intensity in certain regions of the near UV (from 390 nm to 420 nm) could explain the photo-interaction between the aromatic residues. Afterwards, cell culture tests were performed by using mouse NIH3T3 fibroblasts in pursuance of evaluating the preferential tendency of the cells to adhere and proliferate into the different irradiated scaffolds. Following the same goal, we considered to precisely design 3D micro-moulds, by using as an initial imprint a 3D geometry from polylactic acid (PLA) in order to develop a biocompatible 3D RGD-peptide and gelatin-based bio-copolymer scaffold for cellularization. After that, in chapter 3, previously characterized RGD_Cand other peptide derivatives, were studied purposely to behave as a protective band for stem cell therapy. Thereafter, engineered embryonic stem cells (ESCs) were investigated for cell adhesion and short-term proliferation tests in order to evaluate if they maintained a multiplicative potential and a significant viability when seeded in these substrates. Additionally, the investigation of the rheological properties of all these substrates and the further evaluation of functional injectability was studied in view of developing an injectable cell-based therapy. Overall, this thesis will focus on which bioactive peptide-based construct could have a desirable implication in a specific milieu of a damaged tissue so that it will lead up to mimicking the in vivo micro-environment ad hoc. Such 3D cell-based constructs might have a considerable impact in restoring the tissue functionality.
