A STUDY ON THE ACTION AND INTERACTIONS OF HUMAN GROWTH FACTORS FROM BLOOD PLATELETS IN A TOTALLY HOMOLOGOUS CELL CULTURE SYSTEM OF EMBRYONIC FIBROBLASTS. THE EFFECT OF AGING
Abstract
A STUDY ON THE ACTION AND INTERACTIONS OF THE TWO MAJOR GROWTH FACTORS RELEASEDFROM BLOOD PLATELETS-PLATELET-DERIVED GROWTH FACTOR (PDGF) AND TRANSFORMING GROWTH FACTOR-Β (TGF-Β)- IS HERE PRESENTED. WE HAVE DEVELOPED A COMPLETELY HOMOLOGOUS CELL ASSAY SYSTEM, BASED ON HUMAN EMBRYONIC FIBROBLASTS, AND BY USING SPARSE CULTURES WE HAVE SIMULATED IN VITRO THE CONDITIONS PREVAILING IN THE WOUND, WHERE THESE FACTORS ARE RELEASED. IN THIS SYSTEM, PDGF AND TGF-Β STIMULATE CELL PROLIFERATION AND, MOREOVER, THEIR CONCERTED ACTION WAS FOUND TO BE SYNERGISTIC; THE LATTER INDICATES AN IMPORTANT ROLE OF THESE FACTORS DURING TISSUE REGENERATION. FURTHERMORE, THIS SYNERGISM PERSISTS-ALTHOUGH THE NET OVERALL EFFECT GRADUALLY DECREASES-DURING THE IN VITRO AGING OF THE CELLS.
