A study of the ultrastructure of the hepatocyte after the administration of the protein synthesis inhibitors actinomycin or puromycin to newborn rats

Abstract

The effects of the administration of the protein synthesis inhibitors, actinomycin and puromycin, on the electron microscopic appearance of the newborn rat hepatocytes was studied with a special emphasis to the changes related to gluconeogenesis and glycogen breakdown. In this study the following were found. The administration of actinomycin at birth resulted in an inhibition of the normal postnatal development of the peroxisomes (mainly those lacking nucleoids), lysosomes and Golgi apparatus as well as in an inhibition of the hydrolytic breakdown of the glycogen inside the lysosomes. The administration of puromycin resulted in an inhibition of the development of the lysosomes. The results show that the development of the structural features of the metabolic processes of gluconeogenesis and lysosomal glycogen beakdown depend on protein synthesis and that the messenger RNAs for this development are formed after birth.
