DEVELOPMENT OF A RADIOASSAY FOR THE DETERMINATION OF BIOTINIDASE ACTIVITY �IN HUMAN SERUM

Abstract

THE PHD THESIS IS "DEVELOPMENT OF A RADIOMETRIC ASSAY FOR THE DETERMINATION OFBIOTINIDASE ACTIVITY IN HUMAN SERUM". THE CLINICAL SYNDROM OF MULTIPLE CARBOXYLASE DEFICIENCIES ASCRIBED TO BIOTINIDASE DEFICIENCY WHICH APPEARS IN REONATES WITH SYMPTOMS THAT LEADTO DEATH. EARLY DIAGNOSIS OF THE DISEASE IS A LIFE-SAVING PROCEDURE SINCE ALL SYMPTOMS DISAPPEAR AFTER BIOTIN TREATMENT FOR A LIFE TIME AND, THIS, PATIENTS WITH BIOTINIDASE DEFICIENCY LIVE NORMALLY. RADIOIODINATED WITH 125I BIOTIL-TYRAMINE AND ITS COLD COUNTERPART WERE SYNTHESIZED IN ORDER TO BE BIOTINIDASE SUBSTRATE. THE ENZYME KINETICS, WITH THE NEW SUBSTRATE, WERE STUDIED AND THE KM VALUES, V MAXIMUM AND KI (IN THE PRESENT OF THE NATURAL SUBSTRATE BIOCYTIN) WERE DETERMINED. THE METHOD ALSO INCLUDES ARIDIN AS A BINDING PROTEIN, AND POLYETHYLENE GLYCOL AS A SEPARATION REAGENT.
