Η συνδετική ιστόνη LeishH1 της Leishmania έχει μελετηθεί διεξοδικά στο εργαστήριο Μοριακής Παρασιτολογίας του ΕΙΠ. Έχει δειχτεί ότι η επισωμική έκφραση της LeishH1 σε L. major προμαστιγωτά οδηγεί σε υπερέκφραση του μορίου και μειώνει την μολυσματικότητα του παρασίτου in vitro και in vivo (Papageorgiou and Soteriadou, 2002; Smirlis et al., 2006). Στα παράσιτα που υπερεκφράζουν την LeishH1 παρατηρήθηκε in vitro καθυστέρηση στη διαφοροποίηση από προμαστιγωτά σε αμαστιγωτά (Smirlis et al., 2006). Στα πλαίσια της διατριβής αυτής μελετήθηκε εάν οι διαφορές αυτές στη μολυσματικότητα και στην διαφοροποίηση των παρασίτων που υπερεκφράζουν την LeishH1 οφείλονται σε διαφορές στον κυτταρικό τους κύκλο. Τα αποτελέσματα αυτά βρίσκοντα σε συμφωνία με δεδομένα για τις συνδετικές ιστόνες στα μετάζωα, καθώς η υπερέκφραση τους καθυστερεί την αντιγραφή του DNA και την πρόοδο του κυτταρικού κύκλου, που στηρίζουν την ιδέα ότι σημαντικές λειτουργίες της ιστόνης Η1 είναι συντηρημένες κατά τη διάρκεια της εξέλιξης. Εφόσον τα επίπεδα έκφρασης της LeishΗ1 επηρεάζουν τον κυτταρικό κύκλο της Leishmania, το ενδιαφέρον στρέφεται σε μόρια-κλειδιά, που ρυθμίζουν τη λειτουργία της LeishΗ1, κυρίως μέσω μετα-μεταφραστικών τροποιήσεων (π.χ. φωσφορυλίωσης). Η LeishΗ1 περιέχει πιθανά μοτίβα φωσφορυλίωσης από σύμπλοκα κινασών/κυκλινών, τα οποία μπορεί να εμπλέκονται στη ρύθμιση του κυτταρικού κύκλου (Papageorgiou and Soteriadou, 2002). Επίσης, στο πρωτόζωο T. cruzi έχει δειχτεί ότι δύο CDK ομόλογες κινάσες του παρασίτου, οι TzCRK3 και TzCRK1 φωσφορυλιώνουν την Η1 in vitro (da Cunha et al., 2005). Στη Leishmania έχουν μελετηθεί οι CDK ομόλογες πρωτεΐνες CRK1 και CRK3, αλλά δεν έχει δειχτεί ότι φωσφορυλιώνουν την LeishΗ1. Συνεπώς αντικείμενο ενδιαφέροντος αποτελεί η μελέτη κινασών του παρασίτου που πιθανόν φωσφορυλιώνουν τη LeishΗ1. Στα πλαίσια αυτά, μελέτηθηκε η κινάση της συνθετάσης του γλυκογόνου (GSK-3) της Leishmania, σαν ένα πιθανό μόριο που φωσφορυλιώνει την LeishΗ1. Τα τελευταία χρόνια, οι πρωτεϊνικές κινάσες θεωρούνται πιθανά μόρια-στόχοι του παρασίτου και οι ερευνητικές προσπάθειες επικεντρώνονται στη διερεύνηση των υπάρχοντων βιβλιοθηκών αναστολέων κινασών και στη δημιουργία νέων βιβλιοθηκών αναστολέων, που θα στοχεύουν Περίληψη 231 επιλεκτικά τις κινάσες του παρασίτου, χωρίς να προκαλούν βλάβη στον τελικό ξενιστή (Hammarton et al., 2003; Naula et al., 2005). Για το σκοπό αυτό, ταυτοποιήθηκε και χαρακτηρίστηκε η μία από τις δύο ομόλογες ισομορφές της GSK-3 στη L. donovani, που ονομάστηκε LdGSK-3short. Η LdGSK-3short αποτελεί ένα υψηλά συντηρημένο μόριο της Leishmania και εμφανίζει 49% ταυτότητα και 68% ομοιότητα με την ανθρώπινη GSK-3β. Η LdGSK-3short ανιχνεύεται και στις δύο μορφές της Leishmania (προμαστιγωτά και αμαστιγωτά). Τα επίπεδα έκφρασης της δεν διαφέρουν κατά τη διάρκεια της ανάπτυξης των προμαστιγωτών από τη λογαριθμική στη στατική φάση, αλλά μειώνονται περίπου τρεις φορές στα αμαστιγωτά, ένα αποτέλεσμα που βρίσκεται σε συμφωνία με πρόσφατα ευρήματα σχετικά με την έκφραση της LdGSK-3short σε L. donovani αξενικά αμαστιγωτά (Rosenzweig et al., 2008). Επιπλέον, η LdGSK-3short εμφανίζει κυτταροπλασματική εντόπιση σε προμαστιγωτά λογαριθμικής φάσης και πυρηνική εντόπιση σε προμαστιγωτά στατικής φάσης. Σημαντικά είναι επίσης τα ευρήματα ότι: α) η LeishΗ1 είναι υπόστρωμα της LdGSK-3short in vitro και η φωσφορυλίωση της αναστέλλεται από την 5-Me-6-BIO (που όπως αναφέρεται στη συνέχεια αναστέλλει επιλεκτικά την LdGSK-3short) και β) η LeishΗ1 συνεντοπίζεται με την LdGSK-3short στον πυρήνα προμαστιγωτών στατικής φάσης. Περισσότερα πειραματικά δεδομένα χρειάζονται για να αποδειχτεί η in vivo αλληλεπίδραση της LdGSK-3short με την LeishH1. Η διερεύνηση του ρόλου της GSK-3short του παρασίτου, έγινε με τη χρήση χημικών αναστολέων της και γενετικών μελετών (ανασυνδυασμένων παρασίτων που υπερεκφράζουν την κινάση ή την ανενεργή μορφή της). Οι χημικοί αναστολείς που χρησιμοποιήθηκαν, οι ιντιρουμπίνες είναι γνωστό ότι στοχεύουν τις κυκλινο-εξαρτώμενες κινάσες (CDKs) και την κινάση της συνθετάσης του γλυκογόνου (GSK-3) ελέγθηκαν για την αντιλεϊσμανιακή δράση τους. Η 3’ οξίμη της 6-βρώμο-ιντιρουμπίνης (6-BIO), η 3’ ακετοξίμη της 6-βρώμο- ιντιρουμπίνης (6-BIA) και η 3’ οξίμη της 6-βρώμο-5-μέθυλο-ιντιρουμπίνης (5-Me-6-BIO) ήταν οι πιο ισχυροί αναστολείς της ανάπτυξης των L. donovani προμαστιγωτών και αμαστιγωτών (τιμές IC50 ? 1,2 μM). ...

Episomal expression of LeishH1 in L. major promastigotes results in overexpression of this molecule and reduces parasite infectivity in vitro and in vivo (Papageorgiou and Soteriadou, 2002; Smirlis et al., 2006). Interestingly, the reduced infectivity of the overexpressing transfectants was associated with a delay in their differentiation to axenic amastigotes, assessed in vitro (Smirlis et al., 2006). In the context of this study, we investigated whether the reduced infectivity and the delay in differentiation of the parasites over-expressing LeishH1 could be attributed to differences in their cell-cycle progression. The results suggest that over-expression of LeishH1 delays both the G1 to S transition and the progression through the S phase of the cell cycle. The results are in line with data on linker histones in metazoa, since their over-expression delays DNA synthesis and cell-cycle progression, which supports the idea that functional features of LeishH1 are conserved during evolution. Since LeishΗ1 levels of expression affect Leishmania cell cycle, we were also interested in molecules that can regulate LeishΗ1 function, especially through posttranslational modifications (e.g. phosphorylation). LeishΗ1 contains phosphorylation motifs by kinase/cyclin complexes, which might be involved in cell-cycle regulation (Papageorgiou and Soteriadou, 2002). In addition in T. cruzi two CDK homologues, TzCRK3 and TzCRK1 phosphorylate histone H1 in vitro (da Cunha et al., 2005). In Leishmania the two most wellstudied CDK homologues, CRK1 and CRK3, have not been shown to phosphorylate LeishΗ1. Consequently, the study of parasite kinases that possibly phosphorylate LeishΗ1 merits to be investigated. In this context, we studied the leishmanial glycogen synthase kinase-3 (GSK-3) homologue, as a molecule that might phosphorylate LeishΗ1. Recently, parasite kinases are considered promising drug targets and research is focused in the exploitation and expansion of chemical libraries of kinase inhibitors, which will selectively target parasite kinases without damaging the host (Hammarton et al., 2003; Naula et al., 2005). To this end, we identified and characterized one of the two GSK-3 forms in L. donovani, namely LdGSK-3short. LdGSK-3short is a highly conserved molecule in Leishmania species and displays 49% identity and 68% similarity with human GSK-3β. LdGSK-3short is expressed in both parasite stages and its expression pattern was comparable in logarithmic and stationary-phase promastigotes, but it was about 3-fold down-regulated in amastigotes, consistent with recent findings on LdGSK-3short expression in L. donovani axenic amastigotes (Rosenzweig et al., 2008). In addition, LdGSK-3short which had cytosolic and flagellar localization in logarithmic-phase promastigotes, displayed nuclear translocation in stationary-phase promastigotes. Of interest are also the findings that: a) LeishΗ1 is phosphorylated by LdGSK-3short in vitro and the phosphorylation is inhibited by 5-Me-6- BIO (as described below it was found to selectively inhibit LdGSK-3short) and b) LeishΗ1 is co-localized with LdGSK-3short in the nucleus of stationary-phase promastigotes. More data is needed in order to prove the in vivo interaction of LdGSK-3short with LeishH1. The investigation of the biological role of LdGSK-3short in the parasite was performed by the use of chemical inhibitors and genetic studies (overexpressing transfectants). The inhibitors used, indirubins are known inhibitors of mammalian GSK-3 and CDKs (Meijer et al., 2003) and were tested for their antileishmanial activity. Three 6-bromo substituted indirubins, 6-BIO, 6-BIA and 5-Me-6-BIO were powerful inhibitors of both L. donovani promastigote and amastigote growth (IC50 ? 1,2 μM). Promastigote treatment with 5-Me-6-BIO resulted in an accumulation of cells in S and G2/M phases of the cell-cycle and in the induction of apoptosis-like death. 6-BIO induced a dose- and time-dependent accumulation of cells in G2/M and the induction of apoptosis-like death in a lower proportion of promastigotes compared to 5-Me-6-BIO and this death process progressed more slowly. Since 6-bromo substitution on the indirubin backbone enhances the selectivity for mammalian GSK-3 over CDKs (Meijer et al., 2003) we investigated whether 5-Me-6-BIO and 6-BIO target LdGSK-3short and CRK3 (the CDK1 homologue) in vitro. Inhibitor screen assays against LdGSK-3short and CRK3 showed that 5-Me-6-BIO, which is a more potent inhibitor of mammalian GSK-3 over CDK1/Cyclin B displayed an approximately 7-fold selectivity for LdGSK-3short over CRK3. However, 6-BIO was about 7-fold more active towards CRK3 than LdGSK-3short, although it is a mammalian GSK-3 selective inhibitor. ...
