Ο δάκος (Bactrocera oleae Gmelin) είναι το πιο επιβλαβές έντομο της καλλιέργειας της ελιάς στη Μεσόγειο. Η υπερβολική χρήση χημικών εντομοκτόνων για τον έλεγχο του εντόμου και η επακόλουθη ζημιά που προκαλείται στο περιβάλλον και τη δημόσια υγεία είναι ένα σοβαρό πρόβλημα. Πρόσφατα, έχουν γίνει και συνεχίζουν να γίνονται αξιοσημείωτες προσπάθειες σε όλον τον κόσμο, για την ανάπτυξη εναλλακτικών μεθόδων ελέγχου, οι οποίες εκμεταλλεύονται βιολογικούς παράγοντες (φυσικούς εχθρούς, αρπακτικά, βακτήρια, ιούς κ.λπ.) ή βιοχημικούς παράγοντες (ορμόνες, φερομόνες κ.λπ.), με στόχο τον έλεγχο του πληθυσμού του εντόμου. Από όλες αυτές τις μεθόδους τα πιο ενθαρρυντικά αποτελέσματα έχουν ληφθεί με τη χρήση της φερομόνης του ίδιου του εντόμου, είτε με την μέθοδο της μαζικής παγίδευσης (mass trapping), χρησιμοποιώντας παγίδες φερομόνης, είτε διαχέοντας στο περιβάλλον μέσω εξατμιστήρων την απαιτούμενη ποσότητα φερομόνης προκαλώντας παρεμπόδιση των συζεύξεων, μέθοδος που έχει γίνει γνωστή ως μέθοδος απ ...

Πρόσφατα, έχουν γίνει και συνεχίζουν να γίνονται αξιοσημείωτες προσπάθειες σε όλον τον κόσμο, για την ανάπτυξη εναλλακτικών μεθόδων ελέγχου, οι οποίες εκμεταλλεύονται βιολογικούς παράγοντες (φυσικούς εχθρούς, αρπακτικά, βακτήρια, ιούς κ.λπ.) ή βιοχημικούς παράγοντες (ορμόνες, φερομόνες κ.λπ.), με στόχο τον έλεγχο του πληθυσμού του εντόμου. Από όλες αυτές τις μεθόδους τα πιο ενθαρρυντικά αποτελέσματα έχουν ληφθεί με τη χρήση της φερομόνης του ίδιου του εντόμου, είτε με την μέθοδο της μαζικής παγίδευσης (mass trapping), χρησιμοποιώντας παγίδες φερομόνης, είτε διαχέοντας στο περιβάλλον μέσω εξατμιστήρων την απαιτούμενη ποσότητα φερομόνης προκαλώντας παρεμπόδιση των συζεύξεων, μέθοδος που έχει γίνει γνωστή ως μέθοδος αποπροσανατολισμού (matting disruption). Για την αποτελεσματική εφαρμογή των τελευταίων αυτών μεθόδων, απαιτείται μια γρήγορη, εύκολη και αξιόπιστη μέθοδος προσδιορισμού της συγκέντρωσης της φερομόνης που είναι παρούσα στους εξατμιστήρες, στο περιβάλλον ή στα ίδια τα έντομα. Οι ιδιαίτερα μικρές συγκεντρώσεις, στις οποίες εφαρμόζεται η φερομόνη, απαιτούν μια αναλυτική μέθοδο υψηλής ευαισθησίας για τον προσδιορισμό της συγκέντρωσής της σε περιβαλλοντικά δείγματα. Μέχρι σήμερα, τα επίπεδα της φερομόνης του δάκου προσδιορίζονται με αέρια χρωματογραφία (GC) καθώς, όπως όλες οι φερομόνες, είναι μια πολύ πτητική ένωση. Ο προσδιορισμός της ωστόσο, με μια τέτοια μέθοδο ενόργανης ανάλυσης, είναι χρονοβόρος και δύσκολος στην εφαρμογή του, διότι: (α) η συλλογή του δείγματος και η επεξεργασία του είναι δύσκολες, καθώς τα δείγματα πρέπει να είναι ελεύθερα από όλες τις ενώσεις μεγάλης σχετικής μοριακής μάζας που συνυπάρχουν με τη φερομόνη και μπορεί να επιμολύνουν τις στήλες της χρωματογραφίας, (β) κάθε δείγμα μετράται ξεχωριστά και με διαφορετικό πρωτόκολλο, το οποίο εξαρτάται από την προέλευσή του (περιβαλλοντικό ή βιολογικό δείγμα), και (γ) κάθε μέτρηση διαρκεί τουλάχιστον 30 min, ενώ είναι απαραίτητες πολλές επαναλήψεις, ώστε να αποφευχθούν τυχαία σφάλματα. Οι ανοσοαναλυτικές μέθοδοι, οι οποίες βασίζονται στη χρήση εξειδικευμένων αντισωμάτων, είναι οι καταλληλότερες τεχνικές για την ανάλυση μεγάλου αριθμού δειγμάτων σε ελάχιστο χρόνο, με μεγάλη ευαισθησία, επαναληψιμότητα και ακρίβεια. Μέχρι σήμερα, ωστόσο, δεν έχει αναπτυχθεί καμία ανοσοανάλυση για φερομόνη εντόμου, επειδή είναι εξαιρετικά δύσκολο να αναπτυχθούν εξειδικευμένα αντισώματα εναντίον αυτών των ενώσεων, λόγω της μικρής μοριακής μάζας και της ιδιάζουσας, πολλές φορές, δομής τους. Αντικείμενο της παρούσας διατριβής ήταν η ανάπτυξη μιας ανοσοανάλυσης, για τον προσδιορισμό της συγκέντρωσης της φερομόνης του δάκου της ελιάς σε βιολογικά και περιβαλλοντικά δείγματα. Η ανάπτυξη μιας ανοσοανάλυσης για τη φερομόνη του δάκου, που να χαρακτηρίζεται από υψηλή ευαισθησία και ακρίβεια και να είναι ικανή να αναλύει μεγάλο αριθμό δειγμάτων σε μικρό χρονικό διάστημα, θα αποτελούσε πολύτιμο εργαλείο για την επιτυχή εφαρμογή της εναλλακτικής μεθόδου καταπολέμησης του εντόμου. Δύο συνθετικά παράγωγα της φερομόνης του δάκου, το καρβοξυ- παράγωγο (+)-β-[3-(1,7-διοξασπιρο[5.5]ενδεκάνιο προπιονικό οξύ (απτένιο Ι) και το αμινο- παράγωγο (+)-δ-[3-(1,7-διοξασπιρο[5.5]ενδεκάνιο βουτυλαμίνη (απτένιο ΙΙ) χρησιμοποιήθηκαν για την ανάπτυξη της ανοσοανάλυσης της φερομόνης. Το απτένιο Ι συζεύχθηκε στην πρωτεΐνη φορέα keyhole limpet hemocyanin, με τη μέθοδο του καρβοδιιμιδίου, ώστε να παρασκευασθεί κατάλληλο ανοσογόνο, το οποίο στη συνέχεια χρησιμοποιήθηκε επιτυχώς για την ανοσοποίηση κουνελιών καθώς και ορνίθων ωοτοκίας. Η ανοσοποίηση ορνίθων θεωρείται ότι προσφέρει σημαντικά πλεονεκτήματα έναντι της ανοσοποίησης θηλαστικών. Στην περίπτωση των ανοσοποιημένων ορνίθων, οι αντίστοιχες ανοσοσφαιρίνες Υ απομονώθηκαν σε μεγάλες ποσότητες από τον κρόκο των αυγών. .....................

Recently, considerable efforts have been paid worldwide, for the development of alternative control methods, which exploit biological factors (natural enemies, predators, bacteria, viruses, etc) or biochemical factors (hormones, pheromones etc), in order to keep the insect population under control. From all these methods the most promising results are obtained by the use of the insect’s own pheromone. The more widely used technique is the mass trapping, combining a pheromone dispenser and a food attractant on an entomotoxic surface. Although unsuccessfully several applications were made, of the sexual confusion technique, in which pheromone is diffused in the environment at quantities enough to make matting disruption. For the successful application of the above methods, a prerequisite is the ability to measure rapidly and accurately the amount of pheromone which is present in the dispensers, in the environment or in the insects. These analyses will allow to determine the exact amount of synthetic pheromone required, for a more efficient attraction to the traps (mass trapping) or to compete successfully the natural pheromone released by the insects (matting disruption). Until today pheromone levels have been determined by gas chromatography (GC), since the pheromones are volatile compounds of low molecular weight. The determinations are, however, time consuming and difficult to perform because: a) the sample collection and treatment is difficult, since the samples must be free of all the high molecular weight compounds that coexist with the pheromone and may contaminate the chromatography columns, b) every sample is measured separately and with different protocol, depending on its origin (environmental or biological sample), and c) every measurement takes at least 30 min and many repetitions are necessary in order to avoid random errors. The immunoassay methods, which are based on the use of specific antibodies, are the most appropriate techniques for analyzing large number of samples in the minimum time, with high sensitivity, precision and accuracy. Until today no insect pheromone immunoassay has been developed, because it is extremely difficult to raise specific antibodies against these low molecular weight compounds which have often unusual structure. The objective of the present work was to develop an immunoassay for the determination of the olive fruit fly pheromone in biological and environmental samples. The development of an immunoassay for this pheromone, which will guarantee high sensitivity and accuracy and in addition, provide the capability of analyzing many samples in short time, may be an invaluable tool for the successful application of alternative methods against this insect pest. Two synthetic derivatives of the olive fruit fly pheromone, the carboxy- derivative (+)-β-[3-(1,7-dioxaspiro[5.5]undecane] propionic acid (hapten I) and the amine- derivative (+)-δ-[3-(1,7-dioxaspiro[5.5]undecane] butylamine (hapten II) were used in the development of the pheromone ELISA. The hapten I was conjugated to the carrier protein keyhole limpet hemocyanin, via the carbodiimide method, for the preparation of a suitable immunogen and was successfully used to raise polyclonal antibodies by immunization of rabbits as well as of laying hens, which is considered an advantageous alternative to the immunization of mammals. In this latter case, the corresponding immunoglobulins Y were isolated at large quantities from the egg yolk. In the case of immunized rabbits, the corresponding antisera were used. The hapten II, on the other hand, after conjugation to a biotin moiety was used for indirect immobilization onto ELISA microwells which had been pre-coated with the glycoprotein avidin. The immunoassay developed was a competitive biotin-avidin ELISA. According to the assay principle, the biotinylated pheromone (hapten II-biotin) is indirectly immobilized on ELISA microwells pre-coated with avidin. .......................
