Ο μεταγραφικός παράγοντας AP-1 παίζει σημαντικό ρόλο στον έλεγχο των κυτταρικών αποκρίσεων μετά από επίδραση ερεθισμάτων, όπως η ενεργοποιημένη μορφή του RAS. Η μεταγραφική ενεργοποίηση μέσω του AP-1 εξαρτάται από διάφορους συνενεργοποιητές (π.χ. ο p300/CBP) αλλά και από τους μεταγραφικούς παράγοντες TAF (TBP Associated Factors), οι οποίοι περιλαμβάνονται στο σύμπλοκο TFIID. Στην παρούσα μελέτη εξετάσαμε πιθανές αλληλεπιδράσεις μεταξύ πρωτεϊνών AP-1 και TAF με σκοπό να ερευνήσουμε την κοινή ρύθμιση γονιδίων-στόχων τα οποία εμπλέκονται στον καρκίνο του παχέος εντέρου αλλά και στο φαινόμενο της επιθηλιακής προς μεσεγχυματικής μετάβασης (Epithelial-Mesenchymal Transition, ΕΜΤ). Τεκμηριώσαμε δύο νέες αλληλεπιδράσεις ανάμεσα στα μέλη των οικογενειών AP-1 και TAF. Η μελέτη εστιάστηκε στην αλληλεπίδραση των παραγόντων c-Jun και TAF4b. Παράλληλα, η αλληλεπίδραση των παραγόντων c-Jun και TAF4b εμφανίστηκε αυξημένη στα κύτταρα Caco-H, ενώ ο παράγοντας TAF4b βρέθηκε με πειράματα μείωσης της έκφρασής του (siRNA) να παίζει σημαντικό ρόλο στη μεταναστευτική ικανότητα των κυττάρων. Επιπλέον, τα κύτταρα στα οποία πραγματοποιήθηκε μείωση της έκφρασης του παράγοντα TAF4b παρουσίασαν μορφολογικές αλλαγές, σχηματίζοντας προεξοχές, ένα κλασικό χαρακτηριστικό μεσεγχυματικών κυττάρων που παρουσιάζουν αυξημένη ικανότητα μετανάστευσης. Ο παράγοντας TAF4b βρέθηκε επίσης να ρυθμίζει την έκφραση των δύο πρωτεϊνών που απαρτίζουν την ημιδεσμοσωμική Ιντεγκρίνη α6β4 η οποία είναι γνωστό ότι εμπλέκεται στη μεταναστευτική ικανότητα των καρκινικών κυττάρων. Επικεντρώνοντας το ενδιαφέρον μας στην Ιντεγκρίνη α6, η οποία βρέθηκε να ρυθμίζεται και από τον παράγοντα c-Jun αναλύσαμε την ικανότητα πρόσδεσης των παραγόντων TAF4b και c-Jun στον υποκινητή του συγκεκριμένου γονιδίου. Ο παράγοντας TAF4b βρέθηκε να στοχεύει ειδικά γονίδια με αλληλουχίες πρόσδεσης ΑΡ-1, ενώ συγκεκριμένα στο γονίδιο της Ιντεγκρίνης α6, βρέθηκε προσδεμένος στην πιθανή αλληλουχία ΑΡ-1 του υποκινητή της μαζί με τον παράγοντα c-Jun, υποστηρίζοντας την υπόθεση κοινής Περίληψη 10 λειτουργίας τους στη ρύθμιση γονιδίων-στόχων.

A common distribution of the two proteins was identified in different colon cancer cell lines as well as in cell lines that overexpress the activated and oncogenic Ha-RASV12 protein, which have adopted an EMT-like phenotype. Interestingly, the interaction between TAF4b and c-Jun was found enhanced in this EMT-like cell line (Caco-H), while TAF4b was shown to play an important role in the migration ability of cells in experiments where the expression levels of TAF4b were reduced by siRNA treatment of cells. At the same time, after reduction of TAF4b expression levels, cells have undergone an interesting morphological change, forming protrusions, a characteristic of mesenchymal and migrating cells. From the genes implicated in cell adhesion/migration, TAF4b is found to regulate the expression of the hemidesmosomal Integrin α6β4, which is known to play an important role in the migrative ability of the cells. We focused on the Integrin α6, which was at the same time found to be regulated by c-Jun and bears an AP-1 site in its promoter. In an attempt to analyze the binding ability of c-Jun and TAF4b on the AP-1 site of the promoter of Integrin α6, while we had already observed the specific transactivation ability of genes bearing an AP-1 site by TAF4b, we discovered a simultaneous binding of c-Jun and TAF4b on the promoter of Integrin a6. Studying the interdependent binding of TAF4b, c-Jun and other members of the AP-1 family on the promoter of Integrin α6 in different colon cancer cell lines, we came to the conclusion that the regulation of the expression of this specific gene by AP-1 and TAF4b was following a cell-type-specific pattern, while the binding of Summary 12 TAF4b on the promoter of Integrin α6 was in part dependent on c-Jun. Briefly, the abundance of AP-1 family members in each cell line regulates the formation of a specific complex between TAF4b and AP-1 around the promoter of Integrin α6, in order to drive its transcription. During parallel studies: a) The AP-1 site of Vimentin was found to be involved in Ha-Ras induced EMT phenomena through its transcriptional regulation by AP-1 family member FRA-1. b) TAF4b was found localized in the nucleolus of the cell nucleus which was identified to be regulated by the MEK-ERK pathway. Based on this data, an interesting study on a possible regulation of Polymerase I regulated genes by TAF4b will follow in the future. In conclusion, in this study we provide evidence indicating that TAF4b does not only interact with c-Jun in a promoter dependent manner but as well contributes to the initiation of cell-type-specific transcription by interacting with variant AP-1 family members. As until now there has been no evidence indicating an involvement of TAF4b in either cancer or metastasis, we underline the importance of defining the independent roles of specific TFIID subunits as cofactors affecting the outcome of transcription. In addition, this study also provides evidence for a central role of AP-1 transcription factor in regulating genes important for EMT.
