Liposomes as carriers of alkyllysophospholipids and mitomycin c

Abstract

MITOMYCIN C(MMC) IS A POTENT ANTITUMOR DRUG BUT ITS CLINICAL USEFULNESS IS LIMITED BY SERIOUS SIDE EFFECTS.IN THIS STUDY WE TRIED TO INCORPORATE TWO LIPOPHILIC MMC PRODRUGS INTO LIPOSOMES IN ORDER TO REDUCE THE SYSTEMIC TOXICITY OF THE DRUG BY OPTIMISING ITS PHARMACOCINETIC PROPERTIES.THE FORMULATIONS ALSO CONTAINED A SECOND ANTINEOPLASTIC AGENT THE ALKYLLYSOPHOSPHOLIPID Et-18-OMe.THE PARTICLES HAD MEAN DIAMETERS WHICH RANGED BETWEEN 80 AND 123nm.THE PREPARATIONS WERE HOMOGENEOUS AND THEY RETAINED THEIR STABILITY INTERMS OF SIZE AND SIZEDISTIBUTION UPON STORAGE AT 40C.LIPOSOME INTEGRITY DURING INCUBORTION WITH RAT PLASMA WAS EXAMINED AND IT WAS FOUND THAT THE INCORPORATION OF A CERTAIN MMC PRODRUG,HMMC,STABILISED THE STRUCTURE OF THE VESICLES.THE ANTIPROLIFERATIVE ACTIVITY OF LIPOSOMAL HMMC WAS EXAMINED IN VITRO AND IT WAS FOUND THAT THE VECICLES WITH THE LOWEST PERCENTAGE OF HMMC WERE LESS POTENT THAN THE OTHER PREPARATIONS.
