THE ROLE OF LIVER IN THE HOMEOSTASIS OF CSF-1
Abstract
IN THE PRESENT STUDY WE INVESTIGATED THE PHYSIOLOGIC MECHANISM OF CLEARANCE OF CSF-1 (IN MICE C3H/HEJ). THE HALF LIFE OF CSF-1 IS 10 MIN. THE MOST IMPORTANT ORGANS FOR THE CLEARANCE OF CSF-1 ARE THE LIVER AND SPLEEN. THE CSF-1 AFTER RECEPTOR MEDIATED ENDOCYDOSIS, IS DESTROYED IN THE LYSOSOMES OF MACROPHAGES. AUTORADIOGRAPHY AFTER 125I CSF-1 INJECTION IN THE LIVER SHOWED THAT KUPFFER CELLS TAKE UP CSF-1. THE NATURE MACROPHAGES REGULATE, THROUGH A MECHANISM OF NEGATIVE FEEDBACK, THE RATE OF MACROPHAGE PRODUCTION AND THE SERUM CSF-1. AFTER KINETIC STUDIES, IN THE ISOLATED LIVER PERFUSION MODEL WE FOUND THAT THE KINETICS OF UPTAKE OF CSF-1 IN HEPATECTOMIZED LIVERS, AND LIVERS AFTER THE INJECTION OF ENDOTOXIN WAS REDUCED IN COMPARISON WITH NORMAL CONTROLS.
