ELECTRORETINOGRAPHIC CHANGES (ERG) AND ELECTRONIC MICROSCOPY STUDY OF THE RETINA AFTER INTRAVITREAL USE OF GANCICLOVIR
Abstract
THE ELECTRORETINOGRAPHIC CHANGES AND THE RETINAL LESIONS REVEALED BY ELECTRON MICROSCOPY WERE STUDIED IN 20 EYES OF RABBITS AFTER INTRAVITREAL INJECTION OF GANCICLOVIR IN DIFFERENT DOSES RANGING FROM 200-600 ΜG/0.1 ML. GANCICLOVIR WASCLEARLY TOXIC ON THE RETINAL IN DOSES 300-600 ΜG/0.1 ML AS IS SHOWN BY ERG CHANGES. THE PATHOLOGIC RETINAL LESIONS REVEALED BY ELECTRON MICROSCOPY WERE CHARACTERIZED BY HYDROPIC DEGENERATION AND IN PLACES DESTRUCTION OF THE CYTOPLASMSURROUNDING THE NUCLEUS OF THE PHOTORECEPTORS, SWELLING OF THE MITOCHONDRIA AND FRAGMENTATION OF THE CRYSTAL IN SEVERAL OF THEM. IT IS INTERESTING THAT GANCICLOVIR SEEMS TO BE TOXIC EVEN IN THE DOSE OF 200 ΜG/0.1 ML FOUR MONTHS AFTER ITS INTRAVITREAL DELIVERY.
