IDENTIFICATION AND BIOCHEMICAL CHARACTERIZATION OF SECRETED PROTEINS DURING �THE STAGES OF FIBROPLAST PROLIFERATION IN CULTURE
Abstract
CHANGES IN THE COMPONENTS OF THE EXTRACELLULAR MATRIX DURING IN VIVO AGEING ARE SIMILAR TO THE CHANGES OCCURING DURING IN VITRO AGEING. THREE SECRETED POLYPEPTIDES (80, 84 AND 87 KDA) WERE OBSERVED IN EARLY PASSAGE HUMAN FIBROBLAST CULTURES WHILE THEY WERE ABSENT FROM LATE PASSAGE CULTURES. INTERFERON-Γ WAS ALSO FOUND TO SUPPRESS THE SECRETION OF THE THREE POLYPEPTIDES. THE THREEPOLYPEPTIDES WERE ISOLATED. POLYCLONAL AND MONOCLONAL ANTIBODIES WERE PRODUCED AND PROVED THAT ALL THREE SHARE COMMON EPITOPES. IT WAS ALSO SHOWN THAT THE 80 AND 84 KDA POLYPEPTIDES ARE GLYCOSYLATED AND STRUCTURAL SIMILARITIES EXIST BETWEEN THEM. AMINOACID SEQUENCES WERE DETERMINED IN THE N-TERMINALS OF THE 80 AND 84 KDA POLYPEPTIDES AND WERE FOUND IDENTICAL. AN AMINOACID SEQUENCE WITHIN THE 84 KDA POLYPEPTIDE WAS ALSO DETERMINED. THE ABOVE SEQUENCES ARE UNKNOWN TO DATA BANKS. THE STUDY OF THE THREE POLYPEPTIDES IS IMPORTANT TO UNDERSTAND THE EFFECTS OF AGEING ON THE EXTRACEL ...
