Το αντικείμενο της παρούσας διατριβής είναι η in vitro μελέτη της δυνατότητας ανάπτυξης συστημάτων τοπικής μεταφοράς και ελεγχόμενης αποδέσμευσης (DDS) αντικαρκινικών φαρμακευτικών ουσιών χρησιμοποιώντας ως φορείς τους ορυκτές ύλες. Για το σκοπό αυτό η έρευνα προσανατολίστηκε προς δύο κατευθύνσεις. Αρχικά, διερευνήθηκε η ικανότητα τριών διαφορετικών ορυκτών, του υδροξυαπατίτη (HAP), του φωτζεσίτη (FAU) και του μοντμοριλλονίτη (ΜΜΤ), να λειτουργήσουν ως φορείς δύο αντικαρκινικών ουσιών, της υδροχλωρικής γεμσιταβίνης (dFdU.HCl) και της οξαλιπλατίνης (DACH-Pt), προκειμένου να σχεδιασθεί ένα αποτελεσματικό DDS. Γι’ αυτό το λόγο μελετήθηκε ο μηχανισμός και η κινητική προσρόφησης της κάθε φαρμακευτικής ουσίας από υδατικά τους διαλύματα από τα τρία ορυκτά, καθώς και ο μηχανισμός εκρόφησής τους σε προσομοιωμένα υγρά σώματος (SBF) από τα ορυκτά αυτά. Επιπλέον, προκειμένου να διευκρινιστούν οι χημικές αλληλεπιδράσεις των μορίων των φαρμακευτικών ουσιών με το εκάστοτε ορυκτό εφαρμόστηκε η τεχνική της μοριακής προσάραξης. Στη συνέχεια, με στόχο την παρασκευή ενός οστικού εμφυτεύματος το οποίο συγχρόνως δύναται να δρα και ως DDS προς χρήση σε ειδικές περιπτώσεις καρκίνου των πορωδών οστών, εξετάστηκε η δυνατότητα παρασκευής δοκιμίων με βάση τον ΗΑΡ όπου θα συνδυάζονται δύο αντικρουόμενες ιδιότητες, μεγάλο πορώδες (>50 %) με εσωτερικά διασυνδεδεμένους πόρους και συγχρόνως, αυξημένη μηχανική αντοχή στις ανώτερες τιμές που εμφανίζουν τα φυσικά πορώδη οστά (10 – 12 MΡa) προκειμένου να προσομοιάζουν με αυτά ώστε να είναι δυνατή η χρήση του ως εμφυτευμάτων. Επιπλέον, θα πρέπει να φέρουν πόρους διαμέτρου περίπου 200 μm ώστε να επιτυγχάνεται η ενσωμάτωσή τους στον οστικό ιστό και παράλληλα να υπάρχει η δυνατότητα χρήσης τους ως εμφυτεύσιμα DDS. Προκειμένου να ενισχυθεί η μηχανική αντοχή των δοκιμίων ΗΑΡ προστέθηκαν πεφρυγμένα και υαλοποιημένα υλικά (φρίττα Α ή Β) σε διαφορετικά ποσοστά ενώ για την αύξηση του πορώδους προστέθηκαν ως ποροποιητές σφαιρίδια διογκωμένου πολυστυρενίου (ΕPS). Στα δοκίμια που κατασκευάστηκαν με εφαρμογή συμβατικών διεργασιών παραγωγής κεραμικών, προσδιορίστηκαν 5 βασικά τεχνολογικά χαρακτηριστικά (θερμική συστολή, φαινόμενο ειδικό βάρος, υδατοαπορρόφηση, φαινόμενο πορώδες και μηχανική αντοχή σε θλίψη) ενώ με οπτική μικροσκοπία προσδιορίστηκε το μέγεθος των πόρων τους. Η έρευνα ολοκληρώθηκε με τη μελέτη και διαπίστωση της τελικής ικανότητας του παραχθέντος, κατάλληλου πορώδους δοκιμίου ΗΑΡ με αυξημένη μηχανική αντοχή, να λειτουργήσει ως ένα εμφυτεύσιμο DDS φέροντας και αποδίδοντας τοπικά τις αντικαρκινικές φαρμακευτικές ουσίες dFdU.HCl και DACH-Pt.

The overarching goal of the work presented in this dissertation is the development of drug delivery systems (DDS) using minerals as carriers of anticancer chemotherapeutic drugs. The work followed a mainly in vitro approach. The investigation was oriented towards two major directions. First, the behavior of the minerals hydroxyapatite (ΗΑΡ), faujasite (FAU) and montmorillonite (MMT) as carriers of the anticancer drugs gemcitabine hydrochloride (dFdU.HCl) and oxaliplatin (DACH-Pt) for the development of a DDS was investigated. Hence, the production of specific HAP based specimens which would combine two contradicted properties, high porosity (>50 %) with interconnected porous and simultaneously, high compressive strength in the range of the maximum values of cancellous bones (10 – 12 MPa) in order to be simulated and replace the laters as implants was studied extensively. Further, for osteoconduction and osteogenesis purposes as well as for the specimens’ use as implantable DDS, the pore size was chosen to be ~200 μm. The HAP specimens' compressive strength’s improvement was achieved by adding, as fluxing agents, fritts in various percentages while the increment of porosity was achieved by using, as porosifiers, granules of expandable polystyrene (EPS). The specimens were fabricated using traditional ceramics processes. Five technological characteristics of these specimens (thermal construction, bulk density, water adsorption, porosity and compressive strength) were determinated, as well as their pores' size by optical microscopy. Finally, the ability of the porous HAP specimen, with high compressive strength, to act as carrier of the anticancer drugs dFdU.HCl and DACH-Pt in order to be used as implantable DDS was investigated. The prepared specimen was found to be able to carry adequate quantity of the drugs while its technological characteristics fulfilled the aforementioned technical requirements for their apllication.
