ANALYSIS OF RNA STABILITY DURING THE INDUCTION OF DIFFERENTIATION OF ERYTHROLEUKEMIA CELLS (MEL) AND ITS INHIBITION BY DEXAMETHASONE
Abstract
STUDIES OVER THE PAST SEVERAL YEARS HAVE INDICATED THAT INDUCTION OF DIFFERENTIATION OF MURINE ERYTHROLEUKEMIA (MEL) CELLS INTO ORTHOCHROMATOPHILIC NORMOBLASTS, IS CLOSELY ASSOCIATED WITH ALTERATIONS IN THE CYTOPLASMIC LEVEL OF VARIOUS RNA TRANSCRIPTS. SUCH CHANGES HAVE BEEN ATTRIBUTED EITHER TO ALTERATION IN THE RATE OF TRANSCRIPTION OR REPRESSION OF GENES AS WELL AS TO DIFFERENCES IN MECHANISMS THAT GOVERN DIFFERENTIAL MRNA STABILITY DURING THE ENTIRE COURSE OF DIFFERENTIATION OF MEL CELLS ARE NOT AS YET FULLY UNDERSTOOD. RNA STABILITY CAN BE ALTERED BY POST- TRANSCRIPTIONAL STRUCTURAL AND CONFORMATION MODIFICATIONS OF RNATRANSCRIPTS OR BY INTERACTIONS WITH TRANS- ACTING PROTEINS. A FRUITFUL APPROACH TO ADDRESS THESE ISSUES WOULD BE TO TRANSFER RNA INTO MAMMALIAN CELLS AND THEN STUDY THE EFFECT OF THIS RNA TRANSFECTION ON GROWTH, DIFFERENTIATION OF MEL CELLS AND RNA STABILITY WITHIN THE ENVIRONMENT OF UNDIFFERENTIATED AND MATURE CELLS.
