Η TETRAHYMENA PYRIFORMIS ΣΑΝ ΒΙΟΧΗΜΙΚΟ ΣΥΣΤΗΜΑ ΜΕΛΕΤΗΣ ΤΩΝ ΔΡΑΣΕΩΝ ΚΑΙ ΤΗΣ ΠΑΡΑΓΩΓΗΣ ΤΟΥ ΠΑΡΑΓΟΝΤΑ ΕΝΕΡΓΟΠΟΙΗΣΗΣ ΑΙΜΟΠΕΤΑΛΙΩΝ

Abstract

THE UPTAKE OF 1-0-ALKYL-2-(H3)-ACETYL-5N-GLYCERYL-3- PHOSPHORYLALCALINE (AGEPC,PAF) BY TETRAHYMENA PYRIFORMIS WAS INVESTIGATED. TIME COURSE EXPERIMENTS INDICATED THAT MAXIMAL UPTAKE OCCURS AFTER APPROX. 2-8 MIN OF INCUBATION. THERE AFTER, THE AMOUNT OF RADIOACTIVITY RETAINED BY THE CELLS IS DECREASING WITH TIME. THE EFFECT OF SUBSTRATED CONCENTRATION ON THE UPTAKE WAS STUDIED. THERE IS STRONG EVIDENCE THAT AGEPC IS BOUND ON SPECIFIC BINDING SITES. AGEPC (10-6M) RAISES THE LEVEL OF THE PHOSPHATIDIC ACID CONTENT OF THE PROTOZOAN LIPIDS. CHANGES APPEAR TO ALL THE SPECIES OF ACYL CHAINS, WITH MORE PRONOUNCED THAT OF THE PHOSPHATIDYLCHOLINE FRACTION. THERE WAS ALSO A DRASTIC DECREASE OF THE 1-0-ALKYL-5N-GLYCERYL-3-PHOSPHORYLCHOLINE CONTENT OF THE PROTOZOAN LIPIDS. TETRAHYMENA PYRIFORMIS BIOSYNTHESIZES PAF WITHOUT STIMULATION WITH CA++ IONOPHORE A23187 OR ZYMOZAN. TAKE ABSENCE OF CA++ DOES NOT EFFECT PAF PRODUCTION.
